TGF-β signaling links early-life endocrine-disrupting chemicals exposure to suppression of nucleotide excision repair in rat myometrial stem cells

Project description:Environmental exposure to endocrine-disrupting chemicals (EDCs) is associated with uterine fibroids (UFs) development, the most common benign tumors in women of reproductive age. It has been shown that UFs originate from abnormal myometrial stem cells (MMSCs) and there is evidence that defective DNA repair capacity could be involved in the emergence of mutations with implications for tumorigenesis. Moreover, the multifunctional cytokine TGF-β1 has been related to UF progress and some works revealed its connection with certain DNA damage repair pathways. Eker rats develop spontaneous uterine tumors in adulthood, which makes it a useful model for the study of UFs emergence The purpose of this study was to evaluate the impact of endocrine-disrupting chemicals (EDCs exposure on TGF-β1 pathway and nucleotide excision repair (NER) capacity, on Eker rat myometrial stem cells (MMSCs).

Project description:Mass populations of thyrocytes stably expressing wild type RET/PTC1 oncogene or RET/PTC1 carrying Y451F mutation and parental thyrocytes were used for hybridization on Affymetrix HG-U133A and HG-U133B chips. For each cell condition were generated two different targets (indicated as two different samples in the database, i.e. "Parental Thyrocytes" and "Parental Thyrocytes bis")for a total number of six samples. For the data analysis the two samples from the same condition (i.e. Parental thyrocytes) were considered as duplicates.

Cocktail Effect of Endocrine Disrupting Chemicals: Application to Chlorpyrifos in Lavender Essential Oils.

Project description:Chlorpyrifos is a pesticide that is toxic to human health and has been banned for the past decade. Due to its persistent and bioaccumulative properties, chlorpyrifos is still present in soil. Pregnant women can be exposed to chlorpyrifos through drinking water and herbal products, such as essential oils (EOs), resulting in adverse effects to the mother and fetus. Our objective was to evaluate and compare the potential endocrine disrupting effects of chlorpyrifos "free" or in contaminated lavender EO. We studied the release of four hormones and the activation of the P2X7 cell death receptor in human placental JEG-Tox cells as key biomarkers of endocrine toxicity for pregnant women (hPlacentox assay). We observed that "free" chlorpyrifos disrupted placental hormones and activated the P2X7 receptor, whereas chlorpyrifos in lavender EO disrupted only the placental hormones. We confirm that chlorpyrifos can be classified as an endocrine disrupting chemical (EDC) for pregnant women and point out that its endocrine disrupting effect may not be apparent when present in lavender EOs. Our results reveal the existence of specific reverse cocktail effects that may have protective properties against EDCs.

Cell survival and adaptation to chronic endoplasmic reticulum stress in thyrocytes

Project description:The large secretory glycoprotein, thyroglobulin, is the primary translation product of thyroid follicular cells. This difficult-to-fold protein is readily susceptible to structural alterations that render the misfolded thyroglobulin unable to be exported from the endoplasmic reticulum (ER) a known cause of congenital hypothyroidism, with severe, chronic thyrocyte ER stress. Nevertheless, patients with this disease commonly grow a goiter indicative of thyroid cell survival and adaptation. To model this, we have treated PCCl3 thyrocytes with continuous exposure to tunicamcyin (causing an ER stress that can be specifically attributed to thyroglobulin misfolding). In response, PCCl3 cells escape by downregulating expression of Mfsd2a (the tunicamycin transporter). By contrast, following CRISPR/Cas9-mediated deletion of Mfsd2a, PCCl3 cells cannot escape the continuous, chronic effects of high-dose tunicamycin (as demonstrated by persistent accumulation of unglycosylated thyroglobulin); nevertheless the thyrocytes live and grow. A comprehensive proteomic analysis of these cells adapted to chronic ER protein misfolding reveals many hundreds of up-regulated proteins, supporting stimulation of ER chaperones, oxidoreductases, and stress responses as well as lipid biosynthesis pathways. Further, we noted: a) increased phospho-AMP-kinase-B (suggesting upregulated AMPK activity) and decreased phospho-S6 and protein translation (suggesting decreased mTOR activity), consistent with conserved cell survival/adaptation pathways; and b) a suggestion of less differentiated thyrocyte phenotype with decreased PAX8, FOXE1, and TPO protein, as well as a significant decrease of thyroglobulin mRNA levels.
