The Eucalyptus grandis chloroplast proteome: leaf development and seasonal variations

Project description:Chloroplast is a subcellular compartment in which solar radiation is used to generate simple sugar compounds in a complex process named photosynthesis. It is very sensitive to environmental fluctuations and your development is intimately related with the development of the whole plant at different stages. Seasonal climate variations can also drive plant growth and development, favoring a metabolic adaptation of the life cycle with changes in the environment. The characterization of chloroplast proteome dynamics can contribute to a better understanding about the mechanisms involved in leaf development process and seasonal variations in this compartment. Therefore, the goal of this studied was to carried out an exploratory analysis of chloroplast alterations during leaf development and seasonal variations in Eucalyptus grandis chloroplast proteome.

Seasonal Light Hours Modulate Peripheral Clocks and Energy Metabolism in Mice

Project description:With the exception of latitudes close to the equator, seasonal variation in light hours can change dramatically between summer and winter. Yet, investigations into the interplay between energy metabolism and circadian rhythms typically use a 12 h light:12 h dark photoperiod corresponding to light duration at the equator. Here, we hypothesised that altering seasonal photoperiod affects both rhythmicity of peripheral tissue clocks as well as processes involved in energy storage and utilisation. Male mice were housed at one of three photoperiods representing light hours in summer, winter and the equinox. Mice housed at a winter photoperiod exhibited an increase in the amplitude of rhythmic lipid metabolism and a modest reduction in fat mass and liver triglyceride content. Comparing melatonin proficient and deficient mice, we provide evidence that the effect of seasonal light on energy metabolism is largely driven by differences in the rhythmicity of food intake, but not melatonin. Our results show that seasonal light impacts energy metabolism in mice and suggest that these effects are partly driven by modulating the timing of eating. Our work sets a course to integrate seasonal light duration in future circadian biology studies.

Seasonal influenza A virus lineages exhibit divergent abilities to antagonize interferon induction and signaling

Project description:The circulation of seasonal influenza A viruses (IAVs) in humans relies on effective evasion and subversion of the host immune response. While the evolution of seasonal H1N1 and H3N2 viruses to avoid humoral immunity is well characterized, relatively little is known about the evolution of innate immune antagonism phenotypes in these viruses. Numerous studies have established that only a small subset of infected cells are responsible for initiating the type I and type III interferon (IFN) response during IAV infection, emphasizing the importance of single cell studies to accurately characterize the IFN response during infection. We developed a flow cytometry-based method to examine transcriptional changes in IFN and interferon stimulated gene (ISG) expression at the single cell level. We observed that NS segments derived from seasonal H3N2 viruses are more efficient at antagonizing IFN signaling but less effective at suppressing IFN induction, compared to the pdm2009 H1N1 lineage. We compared a collection of NS segments spanning the natural history of the current seasonal IAV lineages, and demonstrate long periods of stability in IFN antagonism potential, punctuated by occasional phenotypic shifts. Altogether, our data reveal significant differences in how seasonal and pandemic H1N1 and H3N2 viruses antagonize the human IFN response at the single cell level.
