Project description:Expression data from xenograft in BALB/c 6-wk-old nude mice with PC3 prostate cancer cells stably expressing PML or a vector control after treatment of the mice with palbociclib (100mg/kg/day diluted in sodium lactate 50mM pH4 given by gavage) during 5 consecutive days PML plays a role in the senescence tumor suppressor response of normal cells to oncogenic stress. PML also mediates therapy-induced senescence in acute promyelocytic leukemia cells after treatment with retinoic acid. However, many tumor cell lines fail to engage a complete senescence response to PML activation. Interestingly, in vivo and in vitro combination of PML overexpression with CDK inhibition induce senescence in PC3 prostate cancer cells. We used microarray to characterize the gene expression profile of PC3 stably expressing PML or a control vector after subcutaneous injection in BALB/c 6-wk-old nude mice and treatment of the mice with palbociclib (100mg/kg/day) during 5 consecutive days..

Effects of oxysterol Oxy186 and Oxy210 on A549 xenograft tumor growth

Project description:To examine the efficacy of oxysterols in inhibiting tumor growth in vivo, we tested Oxy186 and Oxy210 in subcutaneous xenograft tumor models constructed with A549 cells in nude mice. After the tumor size reached an average of ~100 mm3, the compounds were administered daily via oral gavage at 50 mg/kg. We found that Oxy186 started to inhibit tumor growth at day 8, and significantly reduced the tumor volume from day 11 to day18 of treatment, compared to that of vehicle control. Surprisingly, Oxy210 did not show such inhibitory effect.

PIM kinase inhibition in HCC xenograft

Project description:Human HCC cell line, MHCC-97L, was subcutaneously injected to BALB/c nude mice. Two weeks post injection, mice were randomised to receive vehicle or PIM447 (60mg/kg) by oral gavage three times a week for two weeks. Total RNA was extracted from the tumor xenograft (Vehicle, n=2; PIM447, n=2) and subjected to RNA sequencing. Expression profiling of the tumors between vehicle control and PIM447 treated groups was compared.

The regulatory role of lncRNA AC093797.1 in the Hepatocellular carcinoma cell line HCCLM3 in nude mice.

Project description:Purpose: The clinical significance, biological function of AC093797.1 are still unexplored in HCC or any other malignant tumor. In this study, we aimed to investigate the biological function of AC093797.1 in HCC and screen the candidate hub genes and pathways related to hepatocarcinogenesis via RNA-sequencing. Methods: The effects of AC093797.1 on tumor growth in vivo was clarified by nude mice tumor formation experiments. The 5-weeks old female BALB/c nude mice weighing 15-20g were maintained under specific-pathogen-free conditions and randomly divided into OE-AC093797 group and negative control (NC) group (n=5 per group). Then the HCCLM3 cells stably transfected with control vector pcDNA3.1 or the overexpression vector OE-AC093797.1 (5×10^7cells in 100μl) were subcutaneously injected into nude mice. Calculate the volume of the tumors by the following formula: Vtumor= length × width2×π/6. When the tumor grows to about 2000 mm3, the mice were sacrificed after anesthesia and their tumor tissues were isolate. Three mice of each group were used to RNA-sequencing and bioinformatics analysis based on subcutaneous tumor tissue. Results: The “DESeq2” package of R software was used to filtered the differential expressed genes between the OE-AC093797 group and the NC group. The cut-off criteria of differential expressed genes was |log2FoldChange|>1.5 and adj. p value (FDR)<0.05. 710 differentially expressed genes (243 up-regulated genes and 467 down-regulated genes) were screened between groups, which mainly enriched in amino acid metabolism，extracellular matrix structure constituent, cell adhesion molecules cams，signaling to Ras, and signaling to ERKs, etc. via gene ontology, Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis and gene set enrichment analysis (GSEA). Conclusion: AC093797.1 can inhibits tumor group might via reprogram cell metabolism or Extracellular matrix dynamics in HCC.
