Metformin Regulates Metabolic and Non-Metabolic Pathways in Skeletal Muscle and Subcutaneous Adipose Tissues of Older Adults

Project description:Administration of metformin increases healthspan and lifespan in model systems and evidence from clinical trials and observational studies suggests that metformin delays a variety of age-related morbidities. Although metformin has been shown to modulate multiple biological pathways at the cellular level, these pleiotropic effects of metformin on the biology of human aging have not been studied. We studied ~70-year-old participants (n=14), in a randomized, double-blind, placebo-controlled, crossover trial in which they were treated with 6 weeks each of metformin and placebo. Following each treatment period, skeletal muscle and subcutaneous adipose tissue biopsies were obtained, and a mixed-meal challenge test was performed. As expected, metformin therapy lowered 2-hour glucose, insulin AUC, and insulin secretion compared to placebo. Using FDR<0.05, 647 genes were differentially expressed in muscle and 146 genes were differentially expressed in adipose tissue. Both metabolic and non-metabolic pathways were significantly influenced, including pyruvate metabolism and DNA repair in muscle and PPAR & SREBP signaling, mitochondrial fatty acid oxidation and collagen trimerization in adipose. While each tissue had, a signature reflecting its own function, we identified a cascade of predictive upstream transcriptional regulators, including mTORC1, MYC, TNF, TGFß1 and miRNA-29b, that may explain tissue-specific transcriptomic changes in response to metformin treatment. This study provides the first evidence that, in older adults, metformin has metabolic and non-metabolic effects linked to aging. These data can inform the development of biomarkers for the effects of metformin, and potentially other drugs, on key aging pathways. Obesity disrupts cellular cholesterol homeostasis to inhibit the homeostatic clonal expansion of ST2hi VAT Treg cells

Project description:Regulatory T (Treg) cells are highly enriched in the visceral adipose tissue (VAT) to maintain metabolic homeostasis, but they are lost during obesity. Currently, how VAT Treg cells rewire cellular metabolism to support their homeostatic clonal expansion, and whether this process is disrupted in obesity, is unclear. Here, we found that cholesterol metabolism was uniquely upregulated in VAT-, but not other non-lymphoid-tissue Treg cells. Disrupting cholesterol homeostasis (CH) by Treg-specific deletion of Srebf2 led to a specific loss of VAT Treg cells and enhanced obesity-induced systemic metabolic dysfunction. Mechanistically, Srebf2-mediated CH potentiated strong TCR signaling, which specifically promoted the clonal expansion of ST2hi, but not other, VAT Treg subsets. However, long-term high-fat-diet feeding disrupted VAT Treg CH and impaired ST2hi VAT Treg clonal expansion. Restoring Treg CH rescued VAT Treg accumulation in obese mice, suggesting CH modulation could be a potential option for Treg-targeted therapies in obesity-associated metabolic diseases.

Transcriptional profiling of Regulatory T (Treg) cells in obesity

Project description:We reported transcriptional characterization of vTreg53 TCR transgenic Treg cells from splenic and visceral adipose tissue (VAT) in mice fed with normal chow diet (NCD) or high fat diet (HFD) for different durations. We also reported transcriptional profile of VAT Treg cells stimulated by IFNa in vitro. Through this analysis, we found that the reduction of VAT Treg cells during obesity is driven by increased production of IFNa in VAT.

The Effect of Metformin on Maternal and Fetal Outcomes in Pregnant Obese Women (EMPOWaR) trial was a randomised, double-blind, placebo-controlled trial carried out to determine whether exposure to Metformin would affect the offspring birth-weight centile. Obese women exposed to Metformin had increased insulin sensitivity at 36 weeks of pregnancy, but there were no differences in offspring birthweight. We obtained the placentas from these women to determine whether there were differences in expression of genes regulating fetal growth and metabolism. In a complementary study we investigated DNA methylation in the same samples.

Microarray analyses of the effects of prenatal metformin exposure on the gene expression profile of liver, brain and subcutaneous adipose tissue of neonatal mice

Project description:The aim of the study was to characterise the effects of prenatal metformin exposure on a tissue gene expression level in mice. The data consists of two models, regular diet (study I) and high fat diet (study II) model. In both models, metformin (300 mg/kg) or control agent (water) was administered to pregnant female mice from the embryonic day E0.5 untill E17.5. In the high fat diet model, the dams had been on a high fat diet (60% of fat) for one month prior to and during the gestation. The diet was changed to regular diet from E18.5. The pups were sacrificed by decapitation at postnatal day 4 and brain (study I), liver (studies I and II) and subcutaneous adipose tissue (study II) were taken for further microarray analyses. In study I,there are 3 replicates in each group of interest: brain_male_ctrl; brain_male_met; liver_male_ctrl; liver_male_met; liver_female_ctrl; liver_female_met. In study II, there are 6 replicates in each group of interest: liver_male_ctrl; liver_male_met; wat_male_ctrl; wat_male_met.
