Project description:We carried out an time-series transcriptome study of susceptible rice cultivar Nipponbare and resistant cultivar Hui1586 in response to M. oryzae isolate Guy11 at 12, 24, 36 and 48 hpi. In total, 48 samples were subjected to RNA-seq, resulting in 6.8-gigabyte 100-bp paired-end reads per sample on average.

Project description:The rice pathogen, Magnaporthe oryzae, undergoes a complex developmental process leading to formation of an appressorium prior to plant infection. In an effort to better understand phosphoregulation during appressorium development, a mass spectrometry-based study was undertaken. Phosphosites were identified from phosphoproteins from mycelia, conidia, germlings, and appressoria of the wild type and a ckpA mutant. The cyclic-AMP dependent protein kinase A (CPKA) is required for initial perception of the leaf surface, development of functional appressoria, and timely mobilization of storage reserves in conidia, including glycogen and lipid bodies.

Project description:Plants and animals have evolved a first line of defense response to pathogens called innate or basal immunity. While basal defenses in these organisms are well studied, there is almost a complete lack of understanding of such systems in fungal species, and more specifically, how they are able to detect and mount a defense response upon pathogen attack. Hence, the goal of the present study was to understand how fungi respond to biotic stress by assessing the transcriptional profile of the rice blast pathogen, Magnaporthe oryzae, when challenged with the bacterial antagonist Lysobacter enzymogenes. Based on microscopic observations of interactions between M. oryzae and wild-type L. enzymogenes strain C3, we selected early and intermediate stages represented by time-points of 3 and 9 hours post-inoculation, respectively, to evaluate the fungal transcriptome using RNA-seq. For comparative purposes, we also challenged the fungus with L. enzymogenes mutant strain DCA, previously demonstrated to be devoid of antifungal activity. A comparison of transcriptional data from fungal interactions with the wild-type bacterial strain C3 and the mutant strain DCA revealed 463 fungal genes that were down-regulated during attack by C3; of these genes, 100 were also found to be up-regulated during the interaction with DCA. Functional categorization of genes in this suite included those with roles in carbohydrate metabolism, cellular transport and stress response. One gene in this suite belongs to the CFEM-domain class of fungal proteins. Another CFEM class protein called PTH11 has been previously characterized, and we found that a deletion in this gene caused advanced lesion development by C3 compared to its growth on the wild-type fungus. We discuss the characterization of this suite of 100 genes with respect to their role in the fungal defense response.

Project description:Here, we performed comparative transcriptomic studies of ΔMorgs mutant and wild-type strains and found that M. oryzae Aa91 (MoAa91), a homolog of the auxiliary activity family 9 protein (Aa9), was required for surface recognition of M. oryzae. We found that MoAA91 was regulated by the MoMsn2 transcription factor and that its disruption resulted in defects in both appressorium formation on the artificial inductive surface and full virulence of the pathogen. We further showed that MoAa91 was secreted into the apoplast space and was capable of competing with the immune receptor chitin elicitor-binding protein precursor (CEBiP) for chitin binding, thereby suppressing chitin-induced plant immune responses. In summary, we have found that MoAa91 is a novel signaling molecule regulated by RGS and RGS-like proteins and that MoAa91 not only governs appressorium development and virulence but also functions as an effector to suppress host immunity.

Project description:The Ca2+/calcineurin signaling pathway is a central conduit regulating growth, development, and virulence of fungal pathogens infecting plants and human. We have analyzed global gene expression profiles during Ca2+ treatment in the rice blast fungus, Magnaporthe oryzae. An immunosuppressive drug, FK506, and the knock-out mutant of a transcription factor, MoCRZ1, were included to analyze calcineurin- and/or CRZ1-dependent gene expression, respectively. About 1,400 genes were up or down regulated by Ca2+ treatment, while about 200 genes seemed to be up-regulated in a calcineurin/CRZ1-dependent manner. This study analyzes global gene expression dynamics during Ca2+ treatment in the rice blast fungus. We have used Agilent M. grisea 2.0 4×44K Microarrays using a single channel hybridization design. An immunosuppressive drug, FK506, and the knock out mutant of a transcription factor, MoCRZ1, were included to analyze calcineurin- and/or CRZ1-dependent gene expression, respectively. To know the time point at which the gene expression fluctuates most significantly, we treated mycelia with CaCl2 for 0, 15, 30, and 60 min. PMC1 expression level was checked by RT-PCR with the highest expression at 30 min time point. Four biological replicates of wild type and mutant mycelia were harvested after 30 min treatment with chemicals. Pairwise comparisons between Ca2+ treated vs. control and Ca2+ vs. Ca2+/FK506 in wild type (WT), and Ca2+ treated in WT vs. in mutant were conducted.

Project description:Magnaporthe oryzae is notorious for its efficient evolution as a plant pathogen. Analysis of well organised fungal pathogenic mechanisms would enable the scientific understanding to fight against pathogens. Histidine phosphotransferase in fungal systems are good targets for antifungals since they are component of SAPK pathway, which is essential for survival of the fungus. The histidine Phosphotransferase YPD1 in M. oryzae is known to be light responsive and important for both asexual reproduction as well as pathogenicity. In this study, the knock-down (KD) mutant of YPD1 was highly sensitive to cell wall perturbing agents and were highly deficient in laccase expression and activity. A RNA-seq transcriptome screening of KD mutant compared to wild type revealed altered expression of about 146 genes important in biotrophic and necrotrophic phases of the pathogen. About 96 genes were found to be down regulated, of which several plant cell wall degrading genes vital for pathogenicity and fungal cell wall maintenance genes important for growth and fungal development were identified. Most of the genes identified are involved in biotrophic invasion and metabolism. Our results explain the reason for the deficiency of the KD mutant in asexual reproduction as well as pathogenicity. Analysis of differentially expressed genes in the KD YPD1 mutant suggests that YPD1 can be a good target for development of novel antifungal compounds, as YPD1 expression affects genes required for pathogenicity.
