Project description:Sex pheromones rapidly affect endocrine physiology and behaviour, but little is known about their effects on gene expression in the neuroendocrine tissues that mediate olfactory processing. In this study we exposed male goldfish for 6 h to waterborne 17,20betaP (4.3 nM) and PGF2α (3 nM), the main pre-ovulatory and post-ovulatory pheromones, respectively. Both treatments elevated milt volume (P = 0.001). Microarray hybridizations were used to male telencephalon samples following PGF2α treatment and identified 71 unique transcripts that were differentially expressed (q < 5%; 67 up, 4 down). Functional annotation of these regulated genes indicates that PGF2α pheromone exposure affects diverse biological processes including nervous system functions, energy metabolism, cholesterol/lipoprotein transport, translational regulation, transcription and chromatin remodelling, protein processing, cytoskeletal organization, and signalling. By using real-time RT-PCR, we further validated three candidate genes, ependymin-II, calmodulin-A and aldolase C, which exhibited 3 - 5 fold increase in expression following PGF2α exposure. Expression levels of some other genes that are thought to be important for reproduction were also determined using real-time RT-PCR. Expression of sGnRH was increased by PGF2α, but not 17,20betaP, whereas cGnRH expression was increased by 17,20betaP but not PGF2α. In contrast, both pheromones increase the expression of glutamate (GluR2a, NR2A) and gamma-aminobutyric acid (GABAA gamma2) receptor subunit mRNAs. This gene expression data link milt release and rapid modulation of neuronal transcription as part of the response to female sex pheromones.

In addition, two dye swaps were performed for our biological samples for the treatments.

Project description:Diverse animal species use multimodal communication signals to coordinate reproductive behavior. Despite active research in this field, however, the brain mechanisms underlying multimodal communication remain poorly understood. Although anuran amphibians rely on acoustic communication some anurans like humans and many mammalian species, often produce auditory signals accompanied by conspicuous visual signals (e.g. vocal-sac inflation). In this study, we used video playbacks to determine the role of vocal-sac inflation in little torrent frogs (Amolops torrentis). The results showed that auditory cues (i.e. male advertisement calls) as well as visual cues are attractive to female torrent frogs, although the auditory cue was more attractive than the visual cue. Moreover, females preferred simultaneous bimodal cues to unimodal cues. Then we exposed females to blank, visual, auditory and audiovisual stimuli and analyzed whole brain tissue gene expression changes using RNA-seq. The hierarchical clustering of differentially expressed genes (DEGs) showed that samples from the same behavioral stimulus category were more aggregative than those from different behavioral categories nearly forming a cluster, and implying a strong relationship between neurogenomic states and momentarily expressed sexual signals. We also found that the Gene Ontology (GO) terms and KEGG pathways involved in energy metabolism were mostly enriched in blank contrast versus visual, acoustic or audiovisual stimuli, suggesting that brain energy use plays a key role in the perception of these stimuli. Moreover, analyses of immediate early genes (IEGs) showed that only the c-fos gene was differently expressed in response to different stimuli. These results increase our understanding of how neurogenomic states reflect brain mechanisms mediating female preferences for different sexual cues.

We selected approximately 3500 sites highly correlated with age from WGBS data from 9 frogs of 3 different age groups (young, approx. 1 year old; mature, approx. 5 years old; old, approx. 9 years old - GSE221656) and designed probes for the development of cost effective targeted bisulfite sequencing (TBSeq) assay. Here, we present the TBSeq assay in sixteen different frogs as a pilot experiment to validate our previous findings.

Project description:Modes of sexual reproduction in eukaryotic organisms are highly diversified. The human fungal pathogen Candida albicans undergoes a phenotypic switch from the white to the opaque phase in order to become mating-competent. In this study, we report that functionally and morphologically differentiated white and opaque cells show a coordinating behavior in the process of mating. Although white cells are mating-incompetent, they are induced to produce sexual pheromones when treated with opposite pheromones or interacted with opaque cells of an opposite mating type. In a co-culture system, pheromones released by white cells induce opaque cells to form mating projections and thus facilitate both opposite- and same-sex mating of opaque cells. Deletion of genes encoding the pheromone precursor proteins and inactivation of the pheromone response signaling pathway (Ste2-MAPK-Cph1) impair the promoting role of white cells (MTLa) in sexual mating of opaque cells. White and opaque cells communicate via a paracrine pheromone signaling and thus create an environment conducive to sexual mating.
