Project description:The hypothesis was tested that insect meal (IM) as protein source influences intermediary metabolism of growing pigs. To test this, 5-week-old crossbreed pigs were randomly assigned to 3 groups of 10 pigs each with similar body weights (BW) and fed isonitrogenous diets either without (CON) or with 5 % IM (IM5) or 10 % IM (IM10) from Tenebrio molitor L. for 4 weeks and skeletal muscle was analyzed using transcriptomics. Transcriptomics of skeletal muscle revealed a total of 198 transcripts differentially expressed between IM10 and CON.

Identification of peptides and proteins in insect based feed ingredients using LCMSMS

Project description:Identification of peptides and proteins of 18 different insect meal samples derived from the species Hermetia illucens (8), Tenebrio molitor (5), Alphitobius diaperinus (3) and Acheta domesticus (2). Proteins were extracted using two different extraction and solubilization methods. Protein digests were analyzed using an ESI-MS/MS maXis Impact UHR-TOF (Bruker) coupled with an UltiMate 3000 HPLC system (Thermo Scientific) using a 1 mm I.D. x 150 mm reverse phase column (Acclaim PepMap 100 C18, Thermo). Mass spectrometry data (.baf files) were converted using compassXport (Bruker DataAnalysis 4.2) and saved as mzXML files. Proteomics data analysis was conducted using the proteoQC package (version 1.18.1) in R (version 3.4.4) running in RStudio (version 1.0.143).

Effect of replacing soybean meal with Hermetia illucens meal on cecal microbiome, liver transcriptome and plasma metabolome of broilers

Project description:Despite the existence of a number of studies investigating the effect of insect meal on the growth performance of broilers, knowledge about the metabolic effects of insect meal in broilers is still scarce. Thus, the present study investigated the effect of partial replacement of soybean meal with Hermetia illucens (HI) larvae meal on the liver transcriptome, the plasma metabolome and the cecal microbiome in broilers. For the study, 72 male one-day-old Cobb 500 broilers were divided into three groups (n = 12) and fed three different diets with either 0% (HI0), 7.5% (HI7.5) or 15% (HI15) defatted HI meal for 35 d. While body weight (BW) gain, feed intake, and feed:gain ratio did not differ between groups, breast muscle weight, carcass yield and apparent ileal digestibility (AID) of 12 amino acids were higher in group HI15 than in group HI0 (P > 0.05). Indicators of α-diversity (Chao1 and Observed) in the cecal digesta were higher in groups HI15 and HI7.5 than in group HI0 (P < 0.05). The abundance of 5 families and 18 genera, all of which belonged to the Firmicutes phylum, in the cecal digesta differed among groups (P < 0.05). Concentrations of butyric acid, valeric acid and isobutyric acid in the cecal digesta were lower in group HI15 than in the other two groups (P > 0.05), whereas those of total and other short-chain fatty acids were not different between groups. Liver transcriptomics revealed a total of 70 and 61 differentially expressed transcripts between groups HI15 vs. HI0 and between groups HI7.5 vs HI0, respectively, (P > 0.05). Targeted metabolomics identified 138 metabolites, most of which were triglyceride species, being different between the three groups (FDR < 0.05). According to this study, dietary inclusion of HI larvae meal has no detrimental impact but increases breast muscle weight and carcass weight in broilers suggesting that HI larvae meal can be recommended as a sustainable alternative protein source for broilers.

Project description:The sustainable development of modern aquaculture must rely on a significant reduction of the fish meal (FM) used in aquafeed formulations. However, FM substitution with alternative ingredients in diets for carnivorous fish species often showed reduced nutrient absorption, significantly perturbed metabolisms and histological changes at both hepatic and intestinal level. In the present study, adult rainbow trout (Oncorhynchus mykiss) were fed three different experimental aquafeed formulations. A control diet with higher FM content (27.3%) than two test formulations in which fish meal was substituted with two more sustainable and promising alternatives: insect meal (Hermetia illucens larvae=10.1%, FM=11.6%) and poultry by-products meal (PBM=14.8%; FM=11.7%). Combined metabolomics and proteomics analyses of fish liver, together with histological examination of liver and intestine demonstrated that a well balanced formulation of nutrients in the three diets allowed high metabolic compatibility of either substitutions, paving the way for innovative and sustainable use of novel raw materials for the fish feed industry. Results show that the main metabolic pathways of nutrient absorption and catabolism were essentially unaltered by alternative feed ingredients, and also histological alterations were negligible. It is demonstrated that substitution of fish meal with sustainable alternatives does not impact on fish metabolism, given proper efforts are put in fulfilling nutritional requirements of rainbow trout.

Serum metabolomics and proteomics to study the antihyperten-2 sive effect of Tenebrio molitor extracts

Project description:This study aimed at integrating metabolomics and proteomics data for a comprehensive view of the molecular targets of intervention of protein extracts from Tenebrio molitor in treating hypertension. Serum samples from spontaneously hypertensive rats and Wistar Kyoto rats were analyzed using a quantitative metabolomics and label-free proteomics approach based on liquid chromatography coupled with high resolution mass spectrometry (LC-HRMS). Among deregulated metabolites and proteins in hypertensive rats, we found 15 metabolites and 17 proteins that were restored by supplementation with Tenebrio molitor protein extract. The combination of metabolomics and proteomics provided useful data to uncover the molecular targets of intervention and the underlying functional mechanism of Tenebrio molitor protein extract in an animal model such as spontaneously hypertensive rats. The results suggested that Tenebrio molitor supplementation could effectively treat hypertension, partially by regulating proteins and molecules mainly involved in biological pathways associated to angiotensinogen-angiotensin, Serin protease inhibitors, kallikrein–kinin, reactive oxygen scavenging, and lipid peroxidation.
