Project description:This study was conducted to investigate the effects of the rapeseed meal (RM) on two strains of gibel carp (A strain and F strain) and to explore whether two strains show the different molecular mechanisms to the fishmeal (FM) and rapeseed meal (RM). Transcriptomic analysis showed that amino acid metabolism, lipid metabolism and digestive system were three main biological processes affected by RM diet in both strains. The differentially expressed genes between strains were involved in amino acid metabolism, immune responses and lipid metabolism.

Microarray analysis of the Asian seabass (Lates calcarifer) brains

Project description:In this study, we attempt to characterize the transcriptomic profile of the Asian seabass brains collected from the male and female sexes. The objective is to identify genes that show sexually dimorphic expression in the brain of this species. For this experiment, Asian seabass were collected from the Marine Aquaculture Center of the Agri-Food & Veterinary Authority of Singapore. There were no treatments carried out in this experiment. Four brains from adult male seabass (5 years old) with M3-type testis and four brains from adult female seabass (5 years old) with F3-type ovaries were used in this experiment. (Gonads were examined by histology and classified according to sexual maturation status as described by Guiguen and colleagues (Guiguen et al. Environmental Biology of Fishes, 1994)).

Project description:In the present study a 44k oligonucleotide salmonid microarray, qPCR and histology were used to investigate transcriptomic responses in the distal intestine during the first week of oral exposure to soybean meal in order to gain insight into early molecular response mechanisms. Intestinal tissue samples were taken on day one, two, three, five and seven after introduction of a soybean-containing diet and compared to a control group fed fishmeal. Day 1, day 2, day 3, day 5 and day 7 (n=10 per condition) post-soybean meal feeding Atlantic salmon hybridized to common reference and compared against control (day 0 of SBM feeding). Tissue = distal intestine.

Project description:In this study, we attempt to characterize the transcriptomic profile of the Asian seabass gonads at various developmental stages. The protandric Asian seabass or barramundi (Lates calcarifer) typically matures as a male at approximately 2–4 years of age and then changes sex to a female in later years. For this experiment, Asian seabass of several ages were collected from the Marine Aquaculture Center of the Agri-Food & Veterinary Authority of Singapore and from farms around Singapore. There were no treatments carried out in this experiment. The gonads were examined by histology and classified according to sexual maturation status as described by Guiguen and colleagues (Guiguen et al. Environmental Biology of Fishes, 1994). Altogether, we analyzed 22 gonadal samples that could be classified into six different types of gonads.

Effect of replacing soybean meal with Hermetia illucens meal on cecal microbiome, liver transcriptome and plasma metabolome of broilers

Project description:Despite the existence of a number of studies investigating the effect of insect meal on the growth performance of broilers, knowledge about the metabolic effects of insect meal in broilers is still scarce. Thus, the present study investigated the effect of partial replacement of soybean meal with Hermetia illucens (HI) larvae meal on the liver transcriptome, the plasma metabolome and the cecal microbiome in broilers. For the study, 72 male one-day-old Cobb 500 broilers were divided into three groups (n = 12) and fed three different diets with either 0% (HI0), 7.5% (HI7.5) or 15% (HI15) defatted HI meal for 35 d. While body weight (BW) gain, feed intake, and feed:gain ratio did not differ between groups, breast muscle weight, carcass yield and apparent ileal digestibility (AID) of 12 amino acids were higher in group HI15 than in group HI0 (P > 0.05). Indicators of α-diversity (Chao1 and Observed) in the cecal digesta were higher in groups HI15 and HI7.5 than in group HI0 (P < 0.05). The abundance of 5 families and 18 genera, all of which belonged to the Firmicutes phylum, in the cecal digesta differed among groups (P < 0.05). Concentrations of butyric acid, valeric acid and isobutyric acid in the cecal digesta were lower in group HI15 than in the other two groups (P > 0.05), whereas those of total and other short-chain fatty acids were not different between groups. Liver transcriptomics revealed a total of 70 and 61 differentially expressed transcripts between groups HI15 vs. HI0 and between groups HI7.5 vs HI0, respectively, (P > 0.05). Targeted metabolomics identified 138 metabolites, most of which were triglyceride species, being different between the three groups (FDR < 0.05). According to this study, dietary inclusion of HI larvae meal has no detrimental impact but increases breast muscle weight and carcass weight in broilers suggesting that HI larvae meal can be recommended as a sustainable alternative protein source for broilers.
