Identification of Toll-like receptor 4 interacting partner through immunoprecipitation-based chemical cross-linking proteomics

Project description:Toll-like receptor 4 (TLR-4) is a transmembrane protein. Its activation leads to NF-kB signaling pathway and proinflammatory cytokines’ production that are responsible for activating innate immunity system. Here, we design a co-immunoprecipitation based cross-linking proteomics approach to reveal the TLR-4 interactome upon treatment of lipopolysaccharides and statin drug in HA-TLR-4 transfected HEK293 cells. A total of 712 differentially expressed proteins were identified and quantified in this study. Selected candidate proteins, macrophage myristoylated alanine-rich C kinase substrate, and creatine kinase, exclusively identified in the treatment of statin along with cross-linkers, were further validated by immunoblotting.

Comparison of gene expression in macrophages tolerised by different toll-like receptor ligands

Project description:To investigate the similarity of toll-like receptor tolerance in macrophages stimulated with different toll-like receptor ligands we stimulated naïve or tolerant macrophages with ligands for TLR4, TLR2, TLR3 and TLR9. The data identifies a core set of genes that are tolerised by all ligands and genes that show TLR specific patterns.

Subcellular quantitative proteomic analysis of herpes simplex virus type 1 infected HEK 293T cells

Project description:Herpes simplex virus type-1 (HSV-1) is wide-spread dsDNA virus that establishes life-long latency and causes diverse severity of symptoms. In this study, HEK 293T cells with low Toll-like receptor (TLR) and Stimulator of interferon genes protein (STING) expression was infected with HSV-1 and subjected to quantitative proteomic analysis. By using a subcellular fractionation strategy and high-performance mass spectrometry, a total of 5,982 host proteins were quantified, of which 484 proteins were differentially regulated. Bioinformatics analysis indicated that multiple signaling pathways were involved in HSV-1 infection.

Mapping myddosome components in human and murine macrophages.

Project description:Toll-like Receptors (TLRs) are key mediators of immune responses to infection. Upon microbial detection, these receptors activate inflammatory signal transduction pathways that involve IκB kinases, mitogen activated protein kinases, ubiquitin ligases and other adaptor proteins. Current models suggest these signaling proteins operate within functionally distinct multiprotein complexes, which are all activated by a receptor-proximal complex known as the myddosome. The mechanisms that connect the effector protein complexes in the TLR pathways are undefined. To define TLR pathway activities, we engineered macrophages to enable proteomic analysis of the endogenous myddosome constituent MyD88 and its interacting proteins. Proteomics demonstrated that myddosomes contain proteins that act at all stages and regulate all effector responses of the TLR signaling pathways.

DC-SIGN initiates an immature dendritic cell phenotype triggering Rho activation that is utilised by HIV-1

Project description:DC-SIGN is a C-type lectin expressed by dendritic cells (DCs) that binds HIV-1, sequestering it within multivesicular bodies to facilitate transmission to CD4+ T cells. Here we characterize the molecular basis of signalling through DC-SIGN by large-scale gene expression profiling and phosphoproteome analysis. Solitary DC-SIGN activation leads to a phenotypically disparate transcriptional program from Toll-like receptor (TLR) triggering with downregulation of MHC II, CD86, and interferon response genes and with induction of the TLR negative regulator ATF3. Phosphoproteome analysis reveals DC-SIGN signals through the leukemia-associated Rho guanine nucleotide exchange factor (LARG) to induce Rho activity. This LARG activation also occurs on DC HIV exposure and is required for effective HIV viral synapse formation. Taken together HIV mediated DC-SIGN signalling provides a mechanism by which HIV evades the immune response yet induces viral spread. Keywords: Activation state, signalling, Toll-like Receptor (TLR)

qRT-PCR of ex vivo Salmonella enterica in TLR4 minus/plus macrophage background

Project description:A major virulence factor in S. enterica is the pathogenicity island Spi2. The goal of the experiment was to determine the impact of the toll-like receptor 4 (TLR-4) on expression of Spi2 in S. enterica wild type (WT) and triple mutant strain (TMS) with deletions in SPI-1 TTSS, SPI-2 TTSS, flagella. The stain 14028 was used for the experiment. Cells were grown in a TLR-4 wildtype (plus) or deleted (minus) macrophage background. Total RNA was extracted and processed by qRT-PCR. Two strains (WT and TMS) grown under 2 conditions (TLR-4 wildtype (plus) or TLR-4 deleted (minus)) - total 4 samples were analyzed in the study. All PCRs were done in quadruplicates, located on the same PCR plate. For each quadruplicates the median value was calculated and used as a data point for further normalization. A set of controls was selected for all data sets using the following criteria: the data points for the controls should have Cp <40 and be present in all the samples.
