Project description:Accurate timing and anticipation of seasonal changes are required to initiate physiological adaptations over the course of the year such as hibernation, changes in metabolism, fattening and reproductive activity. To achieve this, organisms have evolved complex seasonal timekeeping systems that rely on day length sensing (photoperiodism). The aim of this project is to investigate the role of the circadian clock in photoperiodism within the crucial photoperiod sensing tissue the pars tuberalis.

Transcriptome profiling for floral bud development in reblooming cultivar 'High Noon' of Paeonia suffruticosa

Project description:To examine the mechanisms that control flower development, we sequenced the flower bud transcriptomes of ‘High Noon’, a reblooming cultivar of P. suffruticosa × P. lutea. Both full-length isoforms and RNA-seq were sequenced in 3 floral developmental stages. A total of 15.94 Gb raw data and 457.0 million reads were generated in full-length transcript sequencing and RNA-seq.

ChIP-Seq profiling of ELF3,PRR37 and PHYC in response to photoperiod change at different ZTime in Brachypodium distachyon

Project description:Daylength is an important seasonal cue, and genetic variation in the regulation of photoperiodism has been selected for in adapting wheat and barley to northern latitudes. Despite the importance of this trait, the mechanism by which monocot plants sense photoperiod is not known. We find in Brachypodium distachyon that photoperiodism is mediated by phytochromeC (phyC), a chromophore that acts as a molecular timer and measures the length of the night. The phyC signal is directly communicated to the circadian clock via the activity of EARLY FLOWERING3 (ELF3). ELF3 is a central regulator of photoperiodism, and elf3 mutants display a constitutive long day transcriptome. By coordinating an endogenous timer for night length perception with rhythmical circadian clock controlled gene expression, this mechanism resolves how Brachypodium, and likely other monocots, perceive photoperiod. Using ChIP-Seq, we detected that occupancy of ELF3 at various genomic regions varies in response to photoperiod and ZTime.

Tissue-specific models for stem, leaf, and berry of the Cabernet Sauvignon cultivar were generated from the original model, through the integration of RNA-Seq data. These models have been merged into diel multi-tissue models to study the interactions between tissues at light and dark phases.

Project description:In this project, we profiled the dynamics of proteome during Arabidopsis early embryogenesis using nanoproteomics. In addition, we identified some proteins which may be important during this process. Combining with RNA-seq, we unveiled the relationship between RNA and proteins during Arabidopsis early embryogenesis.
