Shotgun proteomics of chemosensory organs of Varroa destructor mite

Project description:Aims of the project is the identification of soluble chemosensory proteins in the olfactory organs of the honeybee mite Varroa destructor. Different families of soluble proteins acting as carriers for odorants have been identified in hexapods, while very limited information is available for other terrestrial Arthropoda. In mites and ticks olfactory organs are located on the distal part of the forelegs and on the capitulum appendages. These two body tagmata and the second pair of legs as control, have been dissected out of phoretics and reproductives Varroa destructor females and protein extracts have been analysed through shotgun proteomics. Protein identification and relative quantification (Label-free quantification, LFQ) has been performed using MaxQuant (version 1.5.8.3). Among the proteins more abundant in appendages bearing chemosensory organs, considering as a model hexapoda soluble olfactory proteins, we have searched for small secreted proteins whose structure include a hydrophobic binding pocket.

Genetic Adaptation of C. elegans to Environment Changes II: Multigenerational Analysis of Genome-wide Chromatin Remodeling

Project description:Chemical modifications to the tails of histone proteins act as gene regulators that play a pivotal role in adaptive responses to environmental stress. Determining the short and long term kinetics of histone marks is essential for understanding their functions in adaptation. We used Caenorhabditis elegans as a model organism to study the histone modification kinetics in response to environmental stress, taking advantage of their ability to live in both terrestrial and aquatic environments. We investigated the multigenerational genome-wide dynamics of five histone marks (H3K4me3, H3K27me3, H4K20me1, H3K36me1, and H3K9me3) by maintaining P0 animals on terrestrial (agar plates), F1 in aquatic cultures, and F2 back on terrestrial environments. We determined the distributions of histone marks in the gene promoter regions and found that H4K20me1, H3K36me1, and H3K9me3 showed up to eleven-fold differences in density, whereas H3K4me3 and H3K27me3 remained highly constant during adaptation from terrestrial to aquatic environments. Furthermore, we predicted that up to five combinations of histone marks can co-occupy single gene promoters and confirmed the colocalization of these histone marks by structured illumination microscopy. The co-occupancy increases with environment changes and different co-occupancy patterns contribute to variances in gene expressions and thereby presents a supporting evidence for the histone code hypothesis.

Short-term exposure to effluent from bulk drug industries

Project description:The Patancheru area near Hyderabad in India is recognized as a key link in the global supply chain for many bulk drugs. A central treatment plant receives wastewater from about 90 different manufacturers and the resulting complex effluent has contaminated surface, ground and drinking water in the region. Ecotoxicological testing of the effluent has shown adverse effects for several organisms, including aquatic vertebrates. In an attempt to start investigating how exposure to effluent-contaminated water may affect humans and other terrestrial vertebrates, rats were tube-fed effluent. Results from microarray and quantitative polymerase chain reaction assays indicated, however, no marked effects on hepatic gene expression after five days exposure. Neither did a clinical analysis of blood serum constituents used as biomarkers for human disease reveal any significant changes, nor were there any effects on weight gain. Taken together, we could not find evidence for any acute toxicity in the rat; however, we cannot rule out that higher doses of effluent or a longer exposure time may still be associated with risks for terrestrial vertebrates. 10 rats (5 controls and 5 exposed to effluent) were subjected to microarray analysis.
