Project description:To investigate the influence of Aspergillus fumigatus on iron regulation in macrophages, we obtained macrophages in culture from human derived monocytes and co-cultured the monocyte-derived macrophages with Aspergillus conidia at a 1:1 ratio. We collected samples at 0, 2, 4, 6 and 8 hours and extracted RNA. We then performed gene expression profiling analysis using data obtained from RNA-seq of control macrophages and macrophage co-cultured with Aspergillus fumigatus at five time points.
Project description:We report a study conducted to investigate the variation on gene expression of the pathogenic fungus Aspergillus fumigatus upon co-cultivation with the pathogenic bacterium Pseudomonas aeruginosa. The study was conducted by investigating the gene expression variation at different time points (45, 90 and 180 minutes after co-incubation). As control, we used data obtained by cultivating the fungus either without bacteria, or with heat-inactivated Pseudomonas.

Project description:Pseudomonas aeruginosa monocultures and co-cultures with Aspergillus fumigatus during growth in SCFM2 were harvested in the stationary phase of growth (triplicates) by aspirating the contents of the petri-dish well into a 15 ml falcon tube (passed over a 40 µm cell strainer) and combining this with an equal volume of RNA-later. Cultures stored in RNAlater were pelleted, resuspended in 1 mL RNA Bee, and RNA was extracted as described previously.
