Phenotype modulation of VSMCs on patterned substrate

Project description:Loss of contractility and acquisition of an epithelial phenotype of vascular smooth muscle cells (VSMCs) are key events in proliferative vascular pathologies such as atherosclerosis and post-angioplastic restenosis. There is no proper cell culture system allowing VSMC differentiation so that it is difficult to delineate the molecular mechanism responsible for proliferative vasculopathy. We investigated whether a micro-patterned substrate could restore the contractile phenotype of VSMCs in vitro. To induce and maintain the differentiated VSMC phenotype in vitro, we introduced a micro-patterned groove substrate to modulate the morphology and function of VSMCs.

For this purpose the following cell populations were compared: (i) transplanted iPSC-Mφ, (ii) Mφ obtained by in vitro differentiation of murine lineage-negative bone marrow cells (BM-Mφ), (iii) non-differentiated CD45.1 iPSC, (iv) murine alveolar Mφ (AMφ) isolated from the BALF of healthy control mice, and (v) iPSC-Mφ recovered from the transplanted animals two months after (PMT-Mφ).

Comparative study of matrix proteins related to eyes on chiton shell plate

Project description:First, dissect and remove the shell plate of the chiton, then observe the distribution of micro eyes on the shell plate of the chiton under the posture microscope, polish off a part of the back of the shell plate, and then use tools to separate the part with micro eyes and the part without micro eyes. Finally, extract the matrix proteins contained in the two parts through agarose gel electrophoresis, and conduct mass spectrometry analysis of the protein bands to obtain matrix protein data
