Project description:We investigated the contentious role of mitochondrial reactive oxygen species (ROS) on mitochondrial DNA (mtDNA) quality and quantity in female reproductive aging. By conditionally knocking out the Sod2 gene in female mouse germline, we observed increased mitochondrial ROS and decreased oocyte quality, primarily due to impacts on OXPHOS complex II and mtDNA encoded mRNA levels. Interestingly, we found no increased mtDNA mutations, but alterations in mtDNA quantity, indicating the susceptibility of mtDNA to the mitochondrial ROS during reproductive aging. Notably, when we further decreased the basal level of mtDNA quantity by deactivating the mtSSB protein in Sod2 conditional knockout females, we observed an exacerbation of reproductive aging effects. This highlights the crucial role of mtDNA quantity in mitigating the impact of oxidative stress on fertility.

Project description:In order to clarify if there is a tradeoff between quantity and quality of oocytes retrieved after ovarian stimulation, we compared the protein expression of mouse oocytes and embryos with vs without a background of ovarian stimulation using gonadotropins, administered according to the standard protocol in use since more than 50 years (PMID 13185277; PMID 13475597). The data we gathered contribute to a more complete understanding of embryonic gene expression, by taking into account the effect of the commonly used superovulation.

Exposure to copper compromises the maturational competency of porcine oocytes via oxidative stress-induced DNA damage

Project description:Copper (Cu) is not only one of the essential trace elements for animal body, but also an important nutrient component for normal physiology and metabolism of animal reproductive system. Lack or excess of copper will directly or indirectly affect animal reproductive activities. However, the effect of copper on reproductive performance of boars and sows has not been studied and the effect of excessive Copper addition on reproductive performance of sows is even less, and the molecular mechanism is poorly understood. Here, we document that copper has the negative effects on the oocyte maturation and Organelle function. We show that copper exposure perturbs the porcine oocyte meiotic maturation and impair the spindle/chromosome structure, displaying an obviously defective spindle assembly, and abnormal distribution of actin dynamics and cortical granules. In addition, single-cell transcriptome analysis identifies target effectors of copper in porcine oocytes, which was further demonstrated that copper exposure affects the distribution and function of mitochondria, and high ROS levels, DNA damage, and early apoptosis in porcine oocytes. Collectively, we demonstrate that copper exposure causes abnormalities in mitochondrial function and distribution, resulting in increased oxidative stress ROS levels, DNA damage and apoptosis, ultimately leading to decreased quality of porcine oocytes.

Nicotinamide mononucleotide supplementation reverses the declining quality of maternally aged oocytes

Project description:Advanced maternal age, defined as 35 years or older, is associated with a decline in both ovarian reserve and oocyte quality, which leads to the female infertility, pregnancy loss, fetal anomalies, stillbirth, and obstetric complications. At present, the effective approaches to counteract the maternal age-related decay of oocyte quality are still not fully determined. Here, we report that in vivo supplementation of nicotinamide mononucleotide (NMN) efficaciously ameliorates the quality of oocytes from naturally aged mice by recovering nicotinamide adenine dinucleotide (NAD + ) levels in oocytes. NMN supplementation increases the number of antral follicles, ovulated oocytes and matured oocytes from aged mice. Specifically, NMN supplementation maintains the normal spindle/chromosome structure and dynamics of cortical granule component ovastacin to ensure the meiotic competency and fertilization ability of aged oocytes. Moreover, single cell transcriptome analysis shows that the beneficial effect of NMN on the aged oocytes is mediated by the restoration of the mitochondrial function, thereby reducing the accumulated ROS to suppress the occurrence of apoptosis. To sum up, our data reveal that supplementation of NMN is a feasible approach to prevent oocyte quality from advanced maternal age-related deterioration, contributing to improve the reproductive outcome of aged women and the assisted reproductive technology.

Nano-encapsulated senolytics cocktail ameliorates germ cell senescence in female mice

Project description:Low-quality oocytes directly affect fertilization and embryo developmental ability, and further contributes to infertility in women. Dasatinib and quercetin, as a senolytics, has been explored extensively in various age-related diseases. Here, we report that nano-encapsulated senolytics cocktail (D+Q) efficaciously ameliorates the the quantity and quality of follicles and oocytes in vitro and in vivo. D+Q cocktail supplementation reduces the level of ROS in aged oocytes, decreases the frequency of fragmentation, maintains the spindle integrity, rescues mislocalized cortical granules, rescues mitochondrial membrane potential and alleviates DNA damage and apoptosis in vitro. Nano-encapsulated D+Q cocktail effectively ameliorates the fertility deficits in the cyclophosphamide-induced primary ovarian failure (POF) mice model. Moreover, RNA sequencing analysis shows that D+Q cocktail improves the fecundity of POF mice by increasing development gene expression and reduces the senescence-associated secretory phenotype (SASP) accumulation. Taken together, our data show that the D+Q cocktail helps to improve assisted reproductive technology and reproductive outcomes in POF.

The maintenance of oocytes in the mammalian ovary involves extreme protein longevity

Project description:In mammals, oocytes are formed in the female embryo and need to be preserved in the ovary to ensure the viability of the next generation. How oocytes are maintained for decades is unclear. Here, we combined pulse-chase stable isotope labelling coupled with mass spectrometry, single-cell RNA-seq, microscopy and NanoSims to create an atlas of protein homeostasis in mouse oocytes and ovaries over the entire reproductive lifespan. Our results show that protein turnover in the ovary is much slower than in other organs, with hundreds of extremely long-lived proteins across a broad range of complexes and pathways, including mitochondria, ribosomes and the cytoskeleton. We propose that slow protein turnover helps to maintain proteostasis in oocytes and the ovary over long periods, protecting the germline across generations.

Increased expression of BubR1 protects against aneuploidy in aging oocytes and extends reproductive lifespan

Project description:Chromosome aneuploidy increases in oocytes with maternal age, and is considered the leading cause for the increased incidence of infertility, miscarriage, and birth defects. Using mRNA-Sequencing of oocytes from 12 month old mouse versus 3 month young mouse, we identified a spindle assembly checkpoint gene, BubR1, whose expression was significantly decreased. We employed a mRNA microinjection based approach to increase BubR1 expression in aging oocytes. We find that increased expression of BubR1 protects against aneuploidy and chromosome misalignment in aging oocytes. After in vitro fertilization, the embryos derived from BubR1 increased expression aging oocytes exhibited chromosome stability as robust as those of the young ones. Furthermore, following embryo transfer, these embryos showed greatly improved developmental competency, with comparable levels of full-term development to those of the young ones. These results indicate that the decline in oocyte quality may be reversible and could lead to treatments that prolong female fertility. Examination of the effect of maternal aging on the mRNA expression in the mature oocytes of the female mice. Naturally ovulated mature oocytes (MII stage) were collected from 6 young (3 month) and 6 aging (12 month) female mice (3 oocytes per mice, 18 oocytes for each group).
