Project description:Phosphorus is an essential limited nutrient for plant growth and modern agricultural productivity. A comprehensive understanding of how plants respond to phosphate starvation is essential to develop more phosphate-use efficient crops. Here we quantified protein-level responses to phosphate (PO4-) and phosphite (PO3-) in Arabidopsis thaliana suspension cells, relative to a phosphate-starved state using label-free proteomics and phosphoproteomics. Phosphite is similarly sensed and taken up by plants as phosphate but cannot be used as a source of phosphorus. Phosphite is hence a useful tool to delineate between non-specific processes related to sensing and transport, and specific responses to phosphorus nutrition. We find that responses to phosphate and phosphite occur mainly at the level of protein phosphorylation, complemented by limited changes in protein abundance, primarily in protein translation, and phosphate transport and scavenging proteins. Phosphorylation changes in proteins involved in core processes such as translation, RNA splicing and kinase signalling are especially important in phosphate starvation. We also find differential phosphorylation in response to phosphate and phosphite in 69 proteins, including splicing factors, translation factors, the PHT1;4 phosphate transporter and the HAT1 histone acetyltransferase—potential phospho-switches sensing changes in phosphorus nutrition. Our study hence illuminates several new aspects of the phosphate-starvation response and identifies important targets for further investigation and crop improvement.

Project description:A mutant previously isolated from a screen of EMS-mutagenized Arabidopsis lines, per1, showed normal root hair development under control conditions but displayed an inhibited root hair elongation phenotype upon Pi deficiency. Additionally, the per1 mutant exhibited a pleiotropic phenotype under control conditions, resembling Pi-deficient plants in several aspects. Under Pi deficiency, the accumulation of Pi and iron was increased in the mutant when compared to the wild-type. Inhibition of root hair elongation upon growth on low Pi media was reverted by treatment with the Pi analog phosphite, suggesting that the mutant phenotype is not the result of a defect in Pi sensing. Reciprocal grafting experiments revealed that the mutant rootstock is sufficient to cause the phenotype. Transcriptional profiling of per1 and wild-type plants subjected to short-term Pi starvation revealed genes that may be important for the signaling of Pi deficiency. We conclude that UBP14 function is crucial for adapting root development to the prevailing local availability of phosphate. Project description:In order to understand the role of the transcriptional regulator RegX3 in regulating gene expression under phosphate limitation, we performed chromatin immunoprecipitation with RegX3 antibody followed by next generation sequencing using the immunoprecipitate. We observed RegX3 binding at a number of loci including genes which were observed to be differentially regulated during growth in phosphate limited vs. phosphate enriched media, suggesting a global role of RegX3 in regulating gene expression under phosphate starvation.

Project description:The metabolic response of maize source leaves to low nitrogen supply was analyzed in maize seedlings by parallel measurements of transcriptome and metabolome profiling. Inbred lines A188 and B73 were cultivated under controlled growth chamber conditions and supplied with either sufficient (15mM) or limiting (0.15mM) nitrate supply. Leaf lamina material was harvested at day 20 and day 30 after germination with the fifth and sixth leaf representing the main source leaf respectively. Four replicates were collecetd from individual plants for each combination of genotype, growth stage and nitrogen treatment. The leaf material was frozen, homogenised and aliquoted for transcriptome and metabolome analysis. The molecular data was further supplemented by phenotypic characterisation of the maize seedlings under investigation. Limited availability of nitrogen caused strong shifts in the metabolite profile of leaves. The transcriptome was less affected by the nitrogen stress but showed strong genotype and age dependent patterns. Nitrogen starvation initiated the selective down-regulation of processes involved in nitrate reduction and amino acid assimilation; ammonium assimilation related transcripts on the other hand were not influenced. Carbon assimilation related transcripts were characterized by high transcriptional coordination and general down-regulation under low nitrogen conditions. Nitrogen deprivation caused a slight accumulation of starch, but also directed increased amounts of carbohydrates into the cell wall and secondary metabolites. The decrease in N availability also resulted in accumulation of phosphate and by strong down-regulation of genes usually involved in phosphate starvation response, underlining the great importance of phosphate homeostasis control under stress conditions.

Project description:We performed a transcriptomic analysis of Pi starvation responses in Arabidopsis thaliana (Columbia-0) wild type plants under phosphate starvation stress and in plants with altered PHR1(-like) activity, comparing mutants of phr1 and phr1-phl1 grown in phosphate-lacking medium. Results show the central role of PHR1 and functionally redundant members of its family in the control of transcriptional responses to Pi starvation.
