Project description:In this work, we discussed the effects Acanthopanax senticosus Harms (ASE) combined with Gastrodia elata Blume (GEB) in the treatment of CIR injury. In pharmacodynamic studies, found that co-administration of ASE and GEB may improve neuronal injury and prevent neuronal apoptosis by reducing oxidative stress and inflammation, and also help prevent CIR injury. On the basis of our hypothesis, we combined the results of transcriptomic and metabonomic analyses and found that ASE and GEB could prevent cerebral ischemia-reperfusion injury induced by middle cerebral artery occlusion by targeting phenylalanine, pyrimidine, methionine, and sphingolipid metabolism. This study provides the basis for the compatibility and efficacy of ASE and GEB.
Project description:To establish the basis for understanding molecular mechanism of seed germination response to temperature, we analyzed transcriptomes in freshly harvested dormant and dry stored after-ripened seeds. The after-ripened seeds started to show visible germination from 36h after the start of imbibition, and almost all the seeds germinated after 3 days. The freshly harvested seeds stayed dormant by imbibition at 26°C, and germination of the after-ripened seeds was almost completely inhibited at 34°C. Total RNA was prepared from 0 (dry), 6 and 24h imbibed seeds to find regulatory genes of seed dormancy and germination.
Project description:RNAseq from Medicago truncatula early post germination (1 and 5mm radicle seeds) from dissected radicles and cotyledons with and without PEG treatment to identify genes induced during the reinduction of the desiccation tolerance at early germination. Dessication tolerant tissues were radicles from 1mm radicle seeds treated with PEG: R1P, and cotyledons from 1- and 5-mm radicle seeds treated or not with PEG: C1, C5, C1P, C5P and desiccation sensitive tissues were radicles from 1mm radicle seeds (R1) and 5mm radicle seeds (R5).
Project description:Transcriptomic study of the impact of osmopriming on rape seeds (Brassica napus L.; cv 'Libomir') during priming process and after germination. The assays were replicated twice on two independent priming and germination experiments. Seeds were osmoprimed in PEG solution (-1.2 MPa osmotic potential) during 7 days, dried to initial moisture content and then germinated for 7 hours on water. The analysis during different phases of priming procedure (soaking and drying), after whole osmopriming process and germination were done. 10 samples, four condition experiment; non dried primed seeds (Pnd) vs. dry unprimed seeds (UPd) (PEG soaking), non dried primed seeds (Pnd) vs dry primed seeds (Pd) (drying after soaking), dry primed seeds (Pd) vs. dry unprimed seeds (UPd) (full osmopriming process), primed seeds imbibed on water (P7h) vs unprimed seeds imbibed on water (UP7h) (germination after osmopriming). Biological replicates: 2 replicates for comparison PEG soaking, drying after soaking, full osmopriming process and germination after osmopriming.

Transcriptome and translatome are compared for light vs dark for 20h of imbibition. In silico comparison will allow to compare transcriptome and translatome for each type of sample.
