Project description:To understand how different stimulation conditions impacted the maturation of B cells and formation of plasma cells in an ex vivo organoid culture, we preformed RNA-seq comparing the CD19+ B cells and CD138+ plasma cells at different time points during the organoid culture. Two stimulation conditions were used including influenza antigen alone or in combination with the TLR7 agonist R848. Finally, two B cell sources were compared to udnerstand if tissue of origin impacted organoid based B cell differentiation.

Three stimulation conditions were used including H1N1 influenza antigen alone, R848, H1N1 in combination with the TLR7 agonist R848. Additionally, a final set of organoid cultures were stimulated with H1N1 in combination with the TLR7 agonist R848 and at days 16 and 24 inhibitors to BCL6 and EZH2 were added.\

Project description:The aim of the experiment was to investigate the effects of stimulation of human endometrial organoids with estrogen (E2) and progesterone (P4) In humans, the endometrium, the uterine mucosal lining, undergoes dynamic changes throughout the menstrual cycle and in pregnancy. We adapted conditions used to establish human adult stem cell-derived organoid cultures to generate 3D cultures of human endometrium. Unlike other mucosal epithelia, the endometrium responds dramatically to ovarian sex hormones, estrogen (E2) and progesterone (P4), which regulate cyclical proliferation and differentiation of endometrial glands with concomitant dynamic temporal and spatial expression of their receptors, ERalpha and PR.

Comparative nuclear proteomic analysis between undifferentiated and differentiation-initiated mouse spermatogonial stem cell cultures

Project description:Mouse spermatogonial stem cells (SSCs) continuously self-renew on the feeder layers in serum-free culture medium supplemented with glial cell line-derived neurotrophic factor and fibroblast growth factor 2. To identify novel nuclear proteins involved in SSC maintenance, comparative proteomic profiling of nuclear proteins was performed between self-renewing and differentiation-initiated SSCs in culture. The self-renewing SSC cultures were established from C57BL/6 mouse testes. Nuclear fractions from self-renewing SSC cultures treated with ethanol as a vehicle control (spermatogonial stem cells) and differentiation-initiated SSC cultures treated with 0.3 μM retinoic acid for 24 h (spermatogonial progenitor cells) were isolated for proteomic analysis.

Temporal gene expression of human-fetal and glioblastoma stem cell cultures under directed differentiation conditions

Project description:Primary glioblastoma (GBM) cultures vary with respect to differentiation competency. We sought to identify putative transcription factors necessary for the differentiation of GBM cultures. In this dataset, we include expression data obtained from 2 human-fetal neural stem cell (HF-NS) cultures and 2 GBM stem cell (GSC) cultures. We assessed changes in gene expression from 3 timepoints during an in vitro differentiation protocol.
