ScRNA-seq time course of B cell differentiation in organoid cultures

ABSTRACT: To understand how different stimulation conditions impacted the maturation of B cells and formation of plasma cells in an ex vivo organoid culture, we preformed 10X single-cell RNA-seq comparing the CD19+ B cells and CD138+ plasma cells at different time points during the organoid culture. Three stimulation conditions were used including H1N1 influenza antigen alone, R848, H1N1 in combination with the TLR7 agonist R848. Additionally, a final set of organoid cultures were stimulated with H1N1 in combination with the TLR7 agonist R848 and at days 16 and 24 inhibitors to BCL6 and EZH2 were added.\
