Project description:The toxic effects of polystyrene nanoparticles on aquatic ecosystems have been predominantly studied, with a focus on oxidative stress and inflammatory responses. However, this study comprehensively examined the effects of polystyrene nanoparticles on the development of zebrafish embryos using single-cell RNA sequencing analysis, explicitly investigating their effects on erythropoiesis. In vivo experiments were conducted using zebrafish embryos to validate the single-cell RNA sequencing analysis. Exposure to polystyrene nanoparticles resulted in a decrease in the proportion of mature erythrocytes due to an increase in immature erythrocytes during the erythrocyte differentiation process. Additionally, heme synthesis was impaired, leading to a decrease in the proportion of erythrocytes. These findings indicated the toxic effects of polystyrene nanoparticles on hematopoiesis.

Project description:In this study, we attempted to compare among the stress of Y2O3 nanoparticle and YCl3 to yeast cells. Yttrium oxide (Y2O3) nanoparticle is used in widespread applications. It has been reported that Y2O3 nanoparticle showed toxic activities. However, we assumed that the most important toxic factor of nanoparticles was metal ion release. We tried to prove the toxicity of yttrium nanoparticles comes from yttrium ion. Therefore, we used YCl3 as a positive control for evaluation of effects of yttrium ion.

Proteomic dissection of nanoparticle-containing vesicles reveals a major role for a macropinocytic-like uptake mechanism and multivesicular bodies during intracellular nanoparticle trafficking

Project description:Rational design of nanocarriers for drug delivery approaches requires an unbiased knowledge of uptake mechanisms and intracellular trafficking pathways. Here we dissected these processes using a quantitative proteomics approach. We isolated intracellular vesicles containing superparamagnetic iron oxide polystyrene nanoparticles and analyzed their protein composition by label free quantitative mass spectrometry. The proteomic snapshot of organelle marker proteins revealed that an atypical macropinocytic-like mechanism mediated the entry of nanoparticles. We show that the entry mechanism is controlled by actin reorganization, atypical macropinocytic signaling and ADP-ribosylation factor 1. Additionally, our proteomics data demonstrated a central role for multivesicular bodies and multilamellar lysosomes in trafficking and final nanoparticle storage. This was confirmed by confocal microscopy and cryo-TEM measurements. By quantitatively analyzing the protein composition of nanoparticle-containing vesicles, our study clearly defines the routes of nanoparticle entry, intracellular trafficking and the proteomic milieu of a nanoparticle-containing vesicle.

Project description:Recent studies have identified the pervasive presence of nano- and microplastics (NMP) in atmospheric air, leading to inevitable human exposure. Human skin, with an extensive surface area of 1.5-2 m², is constantly exposed to airborne NMP, yet the dermal implications of such exposure remain elusive. Our investigation employed fragmented polystyrene (fPS), a plastic commonly found in the air, to evaluate dermal penetration using various skin models. We utilized pyrolysis-gas chromatography/mass spectrometry (py-GC/MS) to quantify fPS that had penetrated the skin model. Furthermore, we examined the biological alterations in skin keratinocytes consequent to fPS exposure and analyzed the regulation of inflammatory response markers. Our findings reveal deleterious effects on skin keratinocytes, underlining the potential risks posed by short-term fPS exposure. This multifaceted skin model study offers new perspectives on the toxicological impacts of NMP.

Investigation of titanium coated imprinted polystyrene surfaces

Project description:In this study the gene expression differences between two titanium surfaces produced at Maastricht University were investigated. These two surfaces were: flat titanium-coated polystyrene and a titanium-coated polystyrene surface imprinted with a pattern selected from an earlier screening study (Ti1018). This pattern was selected based on the osteoinductive properties observed. As a positive control cells on the flat surface were treated with dexamethasone.

Marker gene/pathway discovery for polystyrene particle toxicity in zebrafish larvae

Project description:We use the zebrafish embryo model to study the innate immune response against polystyrene particles. Therefore, we injected 700nm polystyrene into the yolk at 2 dpf and took samples at 1 and 3 days post injection. This deep sequence study was designed to determine the gene expression profile by polystyrene particle toxicity. RNA was isolated from embryos at 1 and 3 days post injection. Wildtypes zebrafish embryos were micro-injected into the yolk (2dpf) with 1nl of 5mg/ml of 700nm red fluorescent polystyrene particles suspended in PVP (Polyvinylpyrrolidone) (n=3), mock injected with pvp (n=2), or Non-injected as a control (n=3). After injections embryos were transferred into fresh egg water and incubated at 28Â°C. At 1 and 3 days post injection 10 embryos per group were snap-frozen in liquid nitrogen, and total RNA was isolated using TRIZOL reagent.
