Project description:Isoprene, a volatile hydrocarbon, is typically emitted from the leaves and other aboveground plant organs; isoprene emission from roots is not well studied. Given its well-known function in plant growth and defense aboveground, isoprene may also be involved in shaping root physiology to resist belowground stress. We used isoprene-emitting transgenic lines (IE) and a non-emitting empty vector and/or wild type lines (NE) of Arabidopsis to elucidate the roles of isoprene in root physiology and salt stress resistance. We assessed root phenotype and metabolic changes, hormone biosynthesis and signaling, and stress-responses under normal and saline conditions of IE and NE lines. We also analyzed the root transcriptome in the presence or absence of salt stress. IE lines emitted isoprene from roots, which was associated with higher primary root growth, root biomass, and root/shoot biomass ratio under both control and salt stress conditions. Transcriptome data indicated that isoprene altered the expression of key genes involved in hormone metabolism and plant responses to stress factors. Our findings reveal that root constitutive isoprene emission sustains root growth also under salinity by regulating and/or priming hormone biosynthesis and signaling mechanisms, amino acids biosynthesis, and expression of key genes relevant to salt stress defense.

Project description:Isoprene is a C5 volatile organic compound, which can protect aboveground plant tissue from abiotic stress such as short-term high temperatures and accumulation of reactive oxygen species (ROS). Here, we uncover new roles for isoprene in the plant belowground tissues. By analyzing Populus x canescens isoprene synthase (PcISPS) promoter reporter plants, we discovered PcISPS promoter activity in certain regions of the roots including the vascular tissue, the differentiation zone and the root cap. Treatment of roots with auxin or salt increased PcISPS promoter activity at these sites, especially in the developing lateral roots (LR). Transgenic, isoprene non-emitting poplar roots revealed an accumulation of O2 - in the same root regions where PcISPS promoter activity was localized. Absence of isoprene emission, moreover, increased the formation of LRs. Inhibition of NAD(P)H oxidase activity suppressed LR development, suggesting the involvement of ROS in this process. The analysis of the fine root proteome revealed a constitutive shift in the amount of several redox balance, signaling and development related proteins, such as superoxide dismutase, various peroxidases and linoleate 9S-lipoxygenase, in isoprene non-emitting poplar roots. Together our results indicate for isoprene a ROS-related function, eventually co-regulating the plant-internal signaling network and development processes in root tissue. Project description:The use of hybrid-poplar tree plantations as a source for biofuels and biomass production in temperate regions of the Northern Hemisphere has also, unintentionally, increased forest isoprene emissions to the atmosphere. The consequences of increased isoprene emissions include higher rates of tropospheric ozone production and increases in atmospheric aerosol production. Using RNA interference (RNAi) to suppress isoprene emission in several gene insertion events of hybrid-poplars, we show that this trait, which has been assumed as a requisite for the tolerance of abiotic stress, is not required for high rates of woody biomass production, even in extremely hot and dry climates. Biomass production over four years in experimental poplar plantations in Arizona and Oregon was similar among genetic lines that emitted or did not emit significant amounts of isoprene. Lines that had substantially reduced isoprene emission rates also showed decreases in flavonol pigments, which reduce oxidative damage during extremes of abiotic stress; a pattern that would be expected to amplify metabolic dysfunction during abiotic stress. Compensatory increases in the expression of other proteomic components, however, such as those that disable superoxide and other free radicals, and the fact that most biomass is produced during the spring, prior to the hottest and driest part of the growing season, explains the apparent paradox of high biomass production with low isoprene emission. The results of this study provide optimism for designing agroforest plantations of the future that provide high rates of lignocellulose production while eliminating detrimental effects of isoprene emission on atmospheric quality.

Project description:Isoprene is a small lipophilic molecule synthetized at chloroplast level and released into the atmosphere. Isoprene-emitting plants are believed to be better protected against abiotic stresses (i.e. temperature, oxidative stress, drought). The mechanism of action of isoprene is still under debate. In this study we compared the physiological responses and proteomic profile of isoprene-emitting (ISPS) and non-emitting (WT and EV) Arabidopsis thaliana plants subjected to a moderate water stress and in control conditions. Our aim was to investigate if isoprene-emitting plants displayed a different proteomic profile which may help explain how isoprene protects plants from stresses. Only ISPS plants were able to maintain the same photosynthesis and fresh weight in water stress and in control condition. To better understand the molecular mechanism underlying isoprene emission, we performed a LC-MS/MS-based proteomic analysis to explore the changes in protein abundance between WT and EV both under control and moderate water stress conditions. Our data suggest that isoprene exerts its protective mechanism at different levels rather than on a single mode, triggering significant changes in chloroplast protein profiles, but also playing important roles in modulating signaling and hormone pathways and even membrane trafficking.

Project description:Poly- and perfluorinated alkyl substances (PFAS) are a group of persistent organic pollutants. Plants can accumulate PFAS but their effect on plant physiology at the molecular level is not understood yet. We used hydroponically-grown maize plants treated with a combination of eleven different PFAS (each at 100 µg L-1) to investigate their bioaccumulation and effects on the growth, physiology and their impact on the root proteome. From the root proteome analysis, we identified 75 differentially abundant proteins, mostly involved in cellular metabolic and biosynthetic processes, translation and cytoskeletal reorganization. Results were validated using quantitative real-time PCR and further substantiated using amino acid and fatty acid profiling.

Project description:Plant traits, such as root and leaf area, influence how plants interact with their environment and the diverse microbiota living within plants can influence plant morphology and physiology. Here, we explored how three bacterial strains isolated from the Populus root microbiome, influenced plant phenotype. We chose three bacterial strains that differed in predicted metabolic capabilities, plant hormone production and metabolism, and secondary metabolite synthesis. We inoculated each bacterial strain on a single genotype of Populus trichocarpa and measured the response of plant growth related traits (root:shoot, biomass production, root and leaf growth rates) and physiological traits (chlorophyll content, net photosynthesis, net photosynthesis at saturating light-Asat, and saturating CO2-Amax). Overall, we found that bacterial root endophyte infection increased root growth rate up to 184% and leaf growth rate up to 137% relative to non-inoculated control plants, evidence that plants respond to bacteria by modifying morphology. However, endophyte inoculation had no influence on total plant biomass and photosynthetic traits (net photosynthesis, chlorophyll content). In sum, bacterial inoculation did not significantly increase plant carbon fixation and biomass, but their presence altered where and how carbon was being allocated in the plant host.

Project description:Cyanobacteria are phototrophic prokaryotes that can convert inorganic carbon as CO2 into organic carbon compounds at the expense of light energy. In addition, they need only a few inorganic nutrients and can be cultivated in high densities using non-arable land and seawater. This features qualified cyanobacteria as attractive organisms for the production of third generation biofuels as part of the development of future CO2-neutral energy production. Synechocystis sp. PCC 6803 represents one of the most widely used cyanobacterial model strains. On the basis of its available genome sequence and genetic tools, many strains of Synechocystis have been generated that produce different biotechnological products. Efficient isoprene production is an attractive goal, since this compound represents not only an energy-rich biofuel but is also used as chemical feedstock. Here, we report on our attempts to generate isoprene-producing strains of Synechocystis. The cDNA of a codon-optimized plant isoprene synthase (IspS) was cloned under the control of different Synechocystis promoters, which ensure strong constitutive or light-regulated ispS expression. The expression of the ispS gene was quantified by qPCR, whereas the amount of isoprene was quantified using GC-MS. Incubation of our strains at different salt conditions had marked impact on the isoprene production rates. Under low salt conditions, a good correlation was found between ispS expression and isoprene production rate. However, the cultivation of isoprene production strains under salt-supplemented conditions decreased isoprene production despite the fact that ispS expression was salt-stimulated. The characterization of the metabolome of isoprene producing strains indicated that isoprene production might be limited by insufficient precursor levels. Our isoprene production rates under low salt conditions were 2 - 6.5times higher compared to the previous report of Lindberg et al. (2010). These results can be used to guide future attempts establishing the isoprene production with cyanobacterial host systems.
