Mouse CSF Analyses of middle cerebral ischemic/reperfusion (CI/R) injury followed by Buyang Huanwu Decoction

Project description:Ischemic stroke is one of the most common causes of death worldwide and a major cause of acquired disability in adults. Buyang Huanwu decoction (BHD), a traditional Chinese medicine prescription, has long been used clinically for neurological recovery after stroke. Its molecular characteristics and related biomarkers for the therapeutic effect in cerebrospinal fluid (CSF) are yet to be explored. In this project, CSF was obtained from acute ischemic stroke induced mice by a middle cerebral ischemic/reperfusion (CI/R) injury. Proteomic and metabolomic analyses of CSF were performed using LC-MS/MS to define the neuroprotective effect of BHD and the related biomarkers.

The effect of XUESAITONG on gene expression profile after focal cerebral ischemia in rats

Project description:Xuesaitong injection (XST), a standardized patent Chinese medicine of Panax notoginseng roots (Sanqi in Chinese), has long been used for effective prevention and treatment of stroke in China. However, the mechanisms underlying its effects against ischemic stroke are still poorly understood. In this study, we focused on the polypharmacology of XST against ischemic stroke with a XST-regulated stroke network analysis. Male Sprague Dawley rat model of MCAO and reperfusion were administered XST for 7 days while the control group was not treated. Three conditions were compared with three replicates each. These are: (1) sham; (2) model (3) XST treatment. Whole genome microarray analysis was performed using Affymetrix rat Genome 230 2.0 chips.

RNA-seq of non-ischemic heart tissues treated with a traditional Chinese medicine formula (BYD) against myocardial ischemia in Sprague Dawley rats

Project description:The myocardial tissues of the ischemic left ventricle and the peri-ischemic rim (an approximately 1 mm rim of normal-appearing tissues) were removed, and the remaining tissues that consisted of non-ischemic left ventricle were obtained for the transcriptional analysis. The total RNA was extracted from the myocardia from the sham (n = 3), myocardial ischemia model (n = 3), and a traditional Chinese medicine formula treated on myocardial ischemia (n = 3) groups. Beads that contained oligo were used to isolate the poly mRNA from the total RNA and then the rRNA was subsequently removed. The amplified library was sequenced on a HiSeq 2500 sequencing machine (Illumina, San Diego, California, USA) with a 50 bp SE strategy. Using CASAVA software, the images generated by the Illumina sequencers were converted into raw reads by base-calling.

DNA methylation variability in a pilot study group with ischemic stroke

Project description:Elevated plasma homocysteine is an independent risk factor for cardiovascular disease and stroke, however the etiology remains poorly understood. Elevated homocysteine is known to inhibit methyltransferases including DNA methyltransferases, but no methylome-wide analysis of elevated homocysteine has been reported. Peripheral blood genomic DNA methylation in 8 Singaporean-Chinese ischemic stroke patients (4 male, 4 female) with varying homocysteine titer and hypertensive status were profiled using methyl-CpG binding domain (MBD) protein-enriched genome sequencing (MBD-seq) on Illumina Genome Analyzer IIx. A methylome wide screen was undertaken for gender, total plasma homocysteine, hypertension and age. The data show considerable variability within the small cohort, including at genes which are related to one carbon metabolism and cardiovascular disease. Peripheral blood genomic DNA methylation in 8 Singaporean-Chinese ischemic stroke patients (4 male, 4 female) was profiled using methyl-CpG binding domain (MBD) protein-enriched genome sequencing (MBD-seq) on Illumina Genome Analyzer IIx. Methylation parrterns were correlated with homocysteine levels, lypertensive status, gender and age.
