Project description:To determine the complement of Ume6-dependent genes expressed during mitosis and/or meiosis in budding yeast we compared wild-type and<br>ume6 deletion strains using Yeast 2.0 high density oligonucleotide microarrays (GeneChips). Samples were analysed from cells growing in rich medium with fermentable (glucose) and non-fermentable (acetate) carbon sources and from cells undergoing meiosis and spore formation in sporulation medium. Expression data were combined with data from a genome-wide Ume6 DNA binding assay and Ume6-target site prediction to identify the most likely direct target genes of Ume6.

Cross-linking mass spectrometry of yeast mitochondria grown on different carbon sources reveal Ndi1, Min8 and Pet9 associated with the electron transport chain

Project description:Mitochondria derived from Saccharomyces cerevisiae grown on either a non-fermentable (glycerol) or a fermentable (glucose) carbon source were cross-linked with BS3. Additionally, by using a stable-isotope labelled quantitative cross-linking approach, we were able to quantify differences in protein-protein cross-links in mitochondria according to the growth condition. Furthermore, so far uncharacterized yeast proteins were put into biological context based on their cross-linking pattern.

Transcription profiling Saccharomyces cerevisiae xylose metabolism

Project description:In the present study transcriptome and proteome of recombinant, xylose-utilising S. cerevisiae grown in aerobic batch cultures on xylose were compared with glucose-grown cells both in glucose repressed and derepressed states. The aim was to study at genome-wide level how signalling and carbon catabolite repression differed in cells grown on either glucose or xylose. The more detailed knowledge about is xylose sensed as a fermentable carbon source, capable of catabolite repression like glucose, or is it rather recognised as a non-fermentable carbon source is important in achieving understanding for further engineering this yeast for more efficient anaerobic fermentation of xylose. Experiment Overall Design: Three aerobic batch fermentations were carried out both on 50 g l-1 glucose and on 50 g l-1 xylose to compare the yeast transcriptome and proteome of cells growing on xylose with that of glucose repressed and glucose derepressed cells. Samples of the xylose-grown cells were harvested at 72 h from the start of the xylose cultures with 32 g l-1 of residual xylose present. Samples of the glucose repressed cells were harvested at 5 h from the start of the glucose cultures with 37 g l-1 of residual glucose present. Samples of the glucose derepressed cells were harvested at 24 h from the start of the glucose cultures containing no glucose but 13 g l-1 of accumulated ethanol.

Project description:This is genome-scale metabolic model of Hanseniaspora uvarum as the representative species for the clade Saccharomycodaceae. This model was generated through homology search using a fungal pan-GEM largely based on Yeast8 for Saccharomyces cerevisiae, in addition to manual curation. This model has been produced by the Yeast-Species-GEMs project from Sysbio (www.sysbio.se). This is model version 1.0.0 accompanying the publication (DOI: 10.15252/msb.202110427), currently hosted on BioModels Database and identified by MODEL2109130013.
