Project description:The reproductive physiology of yaks differs significantly from that of other cattle breeds due to late sexual maturity, low fecundity and short oestrus time. How to improve the reproductive efficiency of yaks has become the main research content and goal of yak reproduction technology. In this study, we collected blood samples from adult female yaks (4-8 years old) during different reproductive periods, including the period of anestrus (Y-A), estrus (Y-E) and pregnancy (Y-P), and investigated the changes of RNA expression and steroid hormone levels in yaks during different reproductive periods by using RNA-seq and target metabolomics, and screened for the genes and regulatory pathways that were differentially expressed. and related regulatory pathways. DEGs such as PDK4, ALAS2, GLP1R, SLC25A39, PGAP6, FOS, CD36, MMP9 and BCL-6 were identified to play key roles in ovarian function, follicular development, hormone homeostasis and energy metabolism. Functional annotation and enrichment analysis indicated that DEGs were involved in ovarian angiogenesis, hormone synthesis and follicular development. It was found that SLC25A39 may affect glucocorticoid homeostasis and physiological readiness by regulating energy metabolism during anestrus, MARCHF2 and DHEA may be closely related to reproductive hormone fluctuation and system activation during estrus, glucocorticoid down-regulation in pregnancy and maintenance of hormone homeostasis and regulation of immune tolerance by DHEA. The results of this study provide a theoretical basis for improving the reproductive performance of yaks and further analysing the reproductive characteristics of yaks.

Project description:The present study aims at understanding differential gene-expression patterns in the female mouse brain tissues at different stages of reproductive life cycle Methods: Total RNA from brain tissues of 3, 12 and 40 week old female mice was subjected to cDNA and labelled cRNA synthesis and hybridized on 4X44k array. For the same samples, transcriptome sequencing was done followed by qPCR validation of the common genes. Data was analyzed using Gene spring GX 11.5 with MM10 reference along with GSEA and STRING analysis. We primarily focused on understanding gene expression changes in female mice brain which occur during various reproductive life stages viz. 3, 12, 40 weeks which is akin to adolescent, mature and pre-menopausal states.The genes involved in metabolic activity, neurological activity, inflammation and signalling along with genes involved in reproductive regulation show differential expression. The changes observed provide physiological insights in changes in the female brain associated with development, fertility and progressing age.

Project description:IntroductionThe dietary protein level plays a crucial role in maintaining the equilibrium of rumen microbiota in yaks. To explore the association between dietary protein levels, rumen microbiota, and muscle metabolites, we examined the rumen microbiome and muscle metabolome characteristics in yaks subjected to varying dietary protein levels.MethodsIn this study, 36 yaks were randomly assigned to three groups (n = 12 per group): low dietary protein group (LP, 12% protein concentration), medium dietary protein group (MP, 14% protein concentration), and high dietary protein group (HP, 16% protein concentration).Results16S rDNA sequencing revealed that the HP group exhibited the highest Chao1 and Observed_species indices, while the LP group demonstrated the lowest. Shannon and Simpson indices were significantly elevated in the MP group relative to the LP group (P < 0.05). At the genus level, the relative abundance of Christensenellaceae_R-7_group in the HP group was notably greater than that in the LP and MP groups (P < 0.05). Conversely, the relative abundance of Rikenellaceae_RC9_gut_group displayed an increasing tendency with escalating feed protein levels. Muscle metabolism analysis revealed that the content of the metabolite Uric acid was significantly higher in the LP group compared to the MP group (P < 0.05). The content of the metabolite L-(+)-Arabinose was significantly increased in the MP group compared to the HP group (P < 0.05), while the content of D-(-)-Glutamine and L-arginine was significantly reduced in the LP group (P < 0.05). The levels of metabolites 13-HPODE, Decanoylcarnitine, Lauric acid, L-(+)-Arabinose, and Uric acid were significantly elevated in the LP group relative to the HP group (P < 0.05). Furthermore, our observations disclosed correlations between rumen microbes and muscle metabolites. The relative abundance of NK4A214_group was negatively correlated with Orlistat concentration; the relative abundance of Christensenellaceae_R-7_group was positively correlated with D-(-)-Glutamine and L-arginine concentrations.DiscussionOur findings offer a foundation for comprehending the rumen microbiome of yaks subjected to different dietary protein levels and the intimately associated metabolic pathways of the yak muscle metabolome. Elucidating the rumen microbiome and muscle metabolome of yaks may facilitate the determination of dietary protein levels.

We analyzed 25 stages/organs of rice reproductive development including early microsporogenesis stages with 57,381 probe sets, and identified around 26,000 expressed probe sets in each stage. Fine dissection of 25 reproductive stages/organs combined with detailed microarray profiling revealed dramatic, coordinated and finely tuned changes in gene expression. Decrease of expressed genes in the pollen maturation process was observed in a similar way with Arabidopsis and maize. An almost equal number of ab initio predicted genes and cloned genes appeared or disappeared coordinated with developmental stage progression. A large number of organ-/stage-specific genes were identified; notably 2,593 probe sets for developing anther, including 932 probe sets corresponding to ab initio predicted genes. Analysis of cell cycle-related genes revealed that several CDKs, cyclins and components of SCF E3 ubiquitin ligase complexes were expressed specifically in reproductive organs. Cell wall biosynthesis or degradation protein genes and transcription factor genes expressed specifically in reproductive stages were also newly identified. Rice genes homologous to reproduction-related genes in other plants showed expression profiles both consistent and inconsistent with their predicted functions. The rice reproductive expression atlas is likely to be the deepest and most comprehensive dataset available, indispensable for unraveling functions of many specific genes in plant reproductive processes that have not yet been thoroughly analyzed.
