Project description:Methylmercury (MeHg) is an environmental pollutant of global public health concern. MeHg is associated with immune dysfunction but the underlying mechanisms are unclear. The most common route of MeHg exposure is through consumption of fatty fish that contain beneficial n-3 polyunsaturated fatty acids (PUFA) that may protect against MeHg toxicity. To better inform individual costs and benefits of fish consumption, we aimed to identify candidate epigenetic biomarkers of biological responses that reflect MeHg toxicity and PUFA protection.

Network analysis identifies temporal regulation of transcriptomic and physiological responses to early drought perception in Brassica rapa.

Project description:The temporal control of gene expression leads to time of day effects on metabolism and physiology. To identify early transcript level changes occurring in the Brassica rapa oil-type variety R500 during initial drought perception we performed an RNA-seq time course experiment. Leaf tissue was collected from well-watered and drought treated 18 day old plants grown under 14h light at 22°C/10h dark at 18°C every 4h over 2 days with 2 biological replicates at every time point. We collected physiology measurements at every time point and observed dynamic time of day changes in photosynthetic rate, stomatal conductance, non-structural carbohydrates and PSII efficiency. Using a co-expression network approach, phase dependent gene modules were correlated with the physiology data to reveal temporally regulated drought responses. This study provides insight into the transcriptome level responses occurring in the plant in the early stages of drought perception before visible signs of stress occur.

A high glycemic burden drives functional and metabolic alterations of human monocytes in patients with type 1 diabetes

Project description:Diabetes mellitus is associated with serious long-term complications, including increased cardiovascular risk and a higher occurrence of infections. These diabetes-related complications are suggestive of altered responses of the innate immune system. Recent studies have shown that energy metabolism of monocytes is a crucial determinant of their functionality. Here we investigate whether metabolism and function of monocytes are changed in patients with diabetes and aim to identify diabetes-associated factors driving these alterations. Patients with type 1 diabetes (T1D) and healthy age-, sex- and BMI-matched nondiabetic controls were recruited and detailed characteristics related to disease status including diabetes duration and glycemic burden (HbA1c levels) were collected. Monocytes were isolated from peripheral blood to determine immune functionality, metabolic responses and to perform transcriptome analyses. Upon ex vivo stimulation with TLR-4 agonist LPS or TLR-2 agonist P3C, monocytes of patients with T1D secreted lower levels of various cytokines (TNFα, IL-1β and IL-1Ra) and had lower glycolytic rates in comparison to monocytes isolated from matched controls. Stratification based on HbA1c levels revealed that a lower cytokine secretion was coupled to a higher glycolytic rate of monocytes in patients with a higher glycemic burden. Gene expression signatures of circulating monocytes displayed an enhanced inflammatory expression pattern that was associated with this high glycemic burden. A high glycemic burden in patients with T1D promotes expression of inflammatory genes of monocytes in vivo and is coupled to an impaired relationship between metabolism and inflammatory function upon activation ex vivo.

LncRNA/mRNA expression profiling for diabetic and reversal rat retina samples

Project description:We employed LncRNA/mRNA expression profiling as a discovery platform identify aberrantly expressed LncRNAs that fail to reverse back after hyperglycemia is terminated. Using rat model of diabetic retinopathy- metabolic memory, LncRNA array was performed in the retina of rats diabetic for eight months, or in poor glycemic control for four months followed by good glycemic control for four additional months. Expression of selected LncRNAs and their associated genes from these profiles were confirmed by quantitative real-time PCR.
