Epigenetic stabilization of DC and DC precursor classical activation by TNFalpha drives protective T-cell polarization

Project description:We report that TNFalpha mediated dendritic cell priming via alterations in histone 3 lysine 4 trimethylation (H3K4me3) sustains lasting protective Th1/Th17 responses during invasive Cryptococcus neoformans pulmonary infection. Cryptococcus neoformans pulmonary infection results in increased global lung dendritic cell H3K4me3 over naive lung dendritic cells. Blockage of TNFalpha during Cryptococcus neoformans pulmonary infection results in decreased H3K4me3 at sites crucial to protective Th1/Th17 responses.

Project description:We compared the transcriptome of subpopulations of Cryptococcus neoformans cells within the lungs and C. neoformans cells cultured in capsule repressing medium, capsule inducing medium, and capsule inducing medium with the addition of 10% conditioned medium from cells grown in capsule repressing medium. The aim of the study was to identify genes that regulate C. neoformans cell body and capsule size reductions.

Methods: The transcriptome profiles of the wild-type, cir1 mutant and hapX mutant were generated by RNA-seq using Illumina Hiseq, in triplicate. The transcriptome profiles of each mutant was compared with that of the wild-type strain. Results: The iron-responsive transcription factors, Cir1 and HapX, are major regulators for iron acquisition and metabolism in C. neoformans.

Results: The Sterol Regulatory Element Binding Protein, Sre1 is a major regulator for the genes involved in ergosterol biosynthetic pathway and iron acquisition in C. neoformans.
