Transcription profiling of kuk and kur depleted embryos of Drosophila

Project description:Expression profile of Drosophila wild-type embryos in comparison to wild-type embryos injected with kuk dsRNA and embryos from kur germline clones. The stage of the embryos was late stage 5/stage 6/early stage 7 (late cellularisation/early gastrulation before germband extension). Four independent wild-type, two kuk and two kur embryo collections were performed. Total RNA was extracted and amplified. Mutant and respective wildtype samples were hybridized together on INDAC oligo arrays.

D. melanogaster gene expression profiling time course of Snail mutants

Project description:Expression profiling of Drosophila melanogaster halo[AJ] snail[Df(2L)TE116GW11] and halo[AJ] sna[V2] homozygous mutant embryos at two timepoints of embryogenesis. Embryos were manually sorted and assayed at cellularisation stage (stage 5 to early stage 6) and at gastrulation stage (late stage 6 to early stage 8). Homozygous mutant embryos were sorted based on their reduced transparency caused by the recessive mutation of halo[AJ]. Stage-matched halo[AJ] embryos served as wildtype control. Total RNA was extracted, amplified and hybridized to Affymetrix GeneChip Drosophila Genome array version 2. Three biological replicates at each condition were generated.

Project description:Drosophila midline(mid), encodes a T-box transcription factor that is required for cell fate specification and tissue morphogenesis during a series of developmental processes. However, little is known about the target of Mid. To identify the Mid regulated genes, we compared expression profiles of mRNA purified from wild-type and mid mutant embryos. Summary file is yw-mid.CHP. yw and mid mutant embryos were selected at later stage of development for RNA extraction and hybridization on Affymetrix microarrays. We identified mid mutant embryos using CyOact-GFP balancer chromosome.

Gene expression profile of frazzled (fra) mutant Drosophila embryos

Project description:In order to analyze the global changes in gene expression resulting from loss of Fra signaling, we performed a microarray experiment comparing Drosophila embryos containing a loss of function fra[3] mutation to age matched wildtype embryos. RNA extracted from fra[3] mutant vs. wildtype embryos were hybridized to the Affymetrix GeneChip Drosophila Genome 2.0 . We selected Drosophila embryos at stage 13, which marks the onset of Fra expression in the embryonic CNS and the early stages of commissural axon guidance in the ventral nerve cord. RNA was extracted from both fra[3] and wildtype embryos. Hybridization experiments were performed on Affymetrix Drosophila Genome 2.0 microarrays.

Transcription profiling of Drosophila melanogaster homozygous mutant embryos in a timecourse of embryogenesis

Project description:Expression profiling of Drosophila Lmd (lame-duck) homozygous mutant embryos in a timecourse of embryogenesis. Five one-hour timepoints were assayed (5-11 hrs after egg laying). Homozygous mutant embryos were collected using GFP-activated embryo sorting at the same time as stage-matched wildtype controls. Four independent collections were performed at each timepoint. Total RNA was extracted and amplified. Mutant and respective wildtype samples were hybridized together on cDNA arrays.
