Project description:The aim of this project was to evaluate the ploidy of a S. cerevisiae *S. kudriavzevii hybrid in comparison to the lab strain S288C. Other wine yeast have been icluded in the project for the global analysis.
Project description:The thermotolerant TTY23, acid tolerant AT22, and thermo-acid tolerant TAT12 yeast strains, generated by adaptive evolution experiments were grown at 30°C, pH 5.0 in the absence of acetic acid (optimal ancestral conditions). The mRNA of each strain was isolated and changes in expression were compared against the parental strain S288C.
Project description:S288C was transformed with plasmids expressing the GCN5 F221A mutant at varying levels. We sought to examine the global impact on gene expression using the Affymetrix yeast 2.0 arrays. TEF promoter strength varied from 32, 68 and 95% of the wild-type expression. Controls included wild-type with empty & GCN5 null strain with empty plasmid. Strains were grown for approximately 7 dblings in triplicate, followed by a mid-log phase RNA extraction. Whole cell RNA was processed for Affymetrix yeast 2.0 arrays.
