Project description:We have used a Comparative Genomic Hybridization (CGH) microarray to determine the effects of mtDNA depletion using ddC on chromosome stability of Holstein cells that were used as the donor cells for embryo production by Somatic Cell Nuclear Transfer. The results showed that there is no duplications or deletions on chromosomes of mtDNA depleted cells when compared with that of non-depleted cells. In conclusion, depleting mtDNA by using ddC had no negative effect on chromosome stability in the donor cells.

Project description:Implantation is crucial for placental development whose quality will directly impact fetal growth and pregnancy success with possible consequences on post-natal health. We postulated that early perturbations of the conceptus-maternal environment communication may alter the endometrium physiology that could account for the final reproductive outcome. Using cattle as an animal model, we compared gene expression profiles of the endometrial caruncular and intercaruncular areas at implantation in three types of pregnancies, namely artificial insemination (AI), in vitro fertilization with embryo transfer (IVF-ET) or somatic cell nuclear transfer (SCNT). Less than 35% of the differentially regulated genes were found to be common between AI, IVF-ET, and SCNT conditions. Compared to AI, numerous biological functions and several canonical pathways and genes were found to be significantly affected in IVF-ET or SCNT, with a major impact on metabolism and immune function in SCNT. Our data show that endometrium can fine-tune its physiology and could be considered as a biological sensor in response to pregnancy manipulations. Determining the limits of the endometrial plasticity should bring new insights on the contribution of the maternal compartment to the issue of pregnancy. Keywords: Fluorescence Microarray 30 samples

Histone Demethylase-Assisted Somatic Cell Nuclear Transfer Facilitates Derivation of Human Pluripotent Stem Cells

Project description:The extremely low efficiency of human embryonic stem cell (hESC) derivation using somatic cell nuclear transfer (SCNT) limits potential application. Blastocyst formation from human SCNT embryos occurs at a low rate and with only some oocyte donors. We previously showed in mice that reduction of histone H3 lysine 9 trimethylation (H3K9me3) through ectopic expression of the H3K9me3 demethylase Kdm4d greatly improves SCNT embryo development. Here we show that overexpression of a related H3K9me3 demethylase KDM4A improves human SCNT, and that, as in mice, H3K9me3 in the human somatic cell genome is an SCNT reprogramming barrier. Overexpression of KDM4A significantly improves the blastocyst formation rate in human SCNT embryos by facilitating transcriptional reprogramming, allowing derivation of NTESCs from all oocyte donors tested using adult AMD patient somatic nuclei donors. This conserved mechanistic insight has potential applications for improving SCNT in a variety of contexts, including regenerative medicine. Here we perform RNA-seq based transcriptome profiling in human Donor (fibroblast cells), in vitro fertilized embryos at 8-cell stages (IVF_8Cell), somatic cell nuclear transfer embryos at 8-cell stages (SCNT_8Cell), SCNT assisted by KDM4A 8-cell embryos (SCNT_KDM4A_8Cell). Besides, we also perform RNA-seq in Control human ES cells (CTR_hES) and SCNT assisted by KDM4A derived human ES cells (NTK) with duplicates.Â

Mouse ooplasm confers context-specific reprogramming capacity

Project description:Enucleated oocytes have the remarkable ability to reprogram somatic nuclei back to totipotency. Here we investigate genome-scale DNA methylation patterns after nuclear transfer and identify specific targets for DNA demethylation as well as NT specific limitations. We find that the ooplasm can confer similar demethylation patterns in both processes except at some repetitive element classes and that germ-line associated promoters are specifically targeted in NT. Investigating alterations in bovine embryonic pluripotency through scRNA-seq and establishing robust Formative stem cell lines

Project description:Bovine, as one of the most significant domestic animals providing humans with milk and meat, while also serving as bioreactors for producing valuable proteins, pose challenges in establishing embryo-derived stable pluripotent stem cells (PSCs) due to the unclear relationship between embryonic epiblast development and maintenance of stem cell pluripotency and self-renewal. Here, we selected six key stages of bovine embryo development (E5, E6, E7, E10, E12, E14) to track pluripotency changes and their signal pathway dependence using modified single-cell transcription sequencing technology. Based on the remarkable similarity of the dependence of WNT/β-catenin, LIF/STAT3, TGFβ/Smads, and FGF/ERK signaling pathways between bovine and porcine during early embryonic lineages development, we have successfully established bovine embryonic epiblast stem cells (bEpiSCs) using porcine pre-gastrulation epiblast stem cell culture system 3i/LAF. The generated bEpiSCs exhibited consistent pluripotency gene expression levels and maintained clonal morphology, normal karyotype, and proliferative capacity for over 110 passages. Moreover, their high-efficiency teratoma formation potential as well as their ability to differentiate into various cell lineages were demonstrated. At the transcriptome level, bEpiSCs displayed similarities with bovine Formative epiblast cells. We also assessed the application potential of bEpiSCs in targeted differentiation towards muscle cells and as donor cells for somatic cell nuclear transfer (SCNT). In summary, this study establish stable Formative bEpiSCs which hold promise for advancements in cell-cultured meat production, gene-edited cloned cattle generation, and animal breeding.
