Project description:Eight healthy human subjects were enrolled in a 6-day simulated shift work protocol. Blood samples were collected during the two 24-hour measurement periods. Blood samples were collected every 4 hours during both measurement periods. Subjects entered the lab on Day 1. At the start of Day 2, the first 24-hour measurement period was started. Subjects slept according to their habitual sleep/wake schedule, followed by a 16-hour constant posture procedure. On days 3-6, the sleep period was delayed by 10 hours. Following the third night on this schedule, subjects underwent another 24-hour measurement period. During both measurement periods, 7 blood samples were collected and PBMCs were isolated. mRNA was extracted, labelled, and hybridized to microarrays.

Healthy volunteers (ages 22-34, BMI average 25.7) were assigned to a 3 day simulated night-shift schedule (6 male, 1 female) or a simulated day-shift (i.e., control, 4 male, 3 female) schedule. For each group, this was followed by a 24-h constant routine protocol, during which blood was drawn at 3-h intervals to measure plasma lipid profiles using LC-MS/MS.

Project description:Proteomic data from human peripheral blood mononuclear cells (PBMCs). Samples taken over a 24 hour constant light and diet following 3 days of normal day shift cycle or a night shift - circadian misaligned schedule where sleep and wake cycles were reversed. Time points (in clock time) 1, 7, 14, 21 hours; 6 volunteers for each condition. Samples were digested with trypsin, then analyzed by LC-MS/MS. Data was searched with MS-GF+ using PNNL's DMS processing pipeline.

Project description:Shift work misaligns the circadian clock and leads to an increased risk of cancer, cardiovascular diseases, and metabolic disorders. Furthermore, food consumed during the rest phase is a major contributor to this misalignment, as food access restricted to the endogenous active phase, at night, prevents against the adverse effects of shift work on obesity and diabetes in rats. In this study, we aimed to investigate the molecular mechanisms by which shift work and food consumption contribute to an increased risk of cardiovascular disorders. To this end, we used a model of shift work in rats, whereby animals are exposed to a 12:12 light:dark cycle and are forced to be active for eight-hours during their natural rest phase during the day, Monday to Friday for five consecutive weeks. Given the preventive effects of temporal restriction of food intake, a group of shift worker rats with food access restricted to the night was included in addition to controls and shift workers. To gain insight into the molecular underpinnings of shift-work associated cardiovascular pathologies, we performed RNA-Seq analysis and Picrosirius Red staining in rat hearts. Our results show that shift work in rats, regardless of the time of food consumption, have an increase in collagen deposition in the heart. In addition, the expression of many genes encoding key fibrotic pathways was found to be up-regulated in shift worker rats that had their food restricted to the active phase. Altogether, our results suggest that five weeks of shift work in rats is capable of inducing cardiovascular disease through an up-regulation of collagen deposition in the heart.

Project description:Misalignment of the endogenous circadian timing system leads to disruption of physiological rhythms and may contribute to the development of the deleterious health effects associated with night shift work. However, the molecular underpinnings remain to be elucidated. Here, we investigated the effect of a 4-day simulated night shift work protocol on the circadian regulation of the human transcriptome. Repeated blood samples were collected over two 24-hour measurement periods from eight healthy subjects under highly controlled laboratory conditions before and 4 days after a 10-hour delay of their habitual sleep period. RNA was extracted from peripheral blood mononuclear cells to obtain transcriptomic data. Cosinor analysis revealed a marked reduction of significantly rhythmic transcripts in the night shift condition compared with baseline at group and individual levels. Subsequent analysis using a mixed-effects model selection approach indicated that this decrease is mainly due to dampened rhythms rather than to a complete loss of rhythmicity: 73% of transcripts rhythmically expressed at baseline remained rhythmic during the night shift condition with a similar phase relative to habitual bedtimes, but with lower amplitudes. Functional analysis revealed that key biological processes are affected by the night shift protocol, most notably the natural killer cell-mediated immune response and Jun/AP1 and STAT pathways. These results show that 4 days of simulated night shifts leads to a loss in temporal coordination between the human circadian transcriptome and the external environment and impacts biological processes related to the adverse health effects associated to night shift work.

Project description:Study Objectives: Sleep deprivation is highly prevalent and caused by conditions such as night shift work or illnesses like obstructive sleep apnea. Compromised sleep is proposed to play a role in several cardiovascular, immune related and neurodegenerative disorders. We recently published human serum proteome changes after a simulated night shift. This study aimed to further explore changes in the human blood serum after 6h of sleep deprivation at night by proteomics and systems biological databases. Methods: Human blood serum samples from 8 self-declared healthy females were analyzed using mass spectrometry and high-pressure liquid chromatography. Each subject was their own control, and two samples were taken from each subject, the first one after 6h of sleep at night and the second one after 6h of sleep deprivation the following night. Biological databases and bioinformatic software were used for systems biological analyzes and comparative analysis with other published sleep-related datasets. Results: Of 494 proteins, 66 were found to be differentially expressed after 6h of sleep deprivation at night. Functional enrichment analysis revealed associations of these proteins with several biological functions related to the regulation of cellular processes like protein- and ion-binding connected to platelet degranulation and blood coagulation, as well as associations with different curated gene sets. Conclusions: This study presents serum proteomic changes after 6h of sleep deprivation, supports previous findings that only 6h of sleep deprivation affects several biological processes and revealed a molecular signature of protein changes related to pathological conditions like altered coagulation and platelet function, impaired lipid and immune function and cancer. Project description:A majority of night shift workers have their circadian rhythms misaligned to their atypical schedule. While bright light exposure at night is known to reset the human central circadian clock, the behavior of peripheral clocks under conditions of shift work is more elusive. The aim of the present study was to quantify the resetting effects of bright light exposure on both central (plasma cortisol and melatonin) and peripheral clocks markers (clock gene expression in peripheral blood mononuclear cells, PBMCs) in subjects living at night. Eighteen healthy subjects were enrolled to either a control (dim light) or a bright light group. Blood was sampled at baseline and on the 4th day of simulated night shift. In response to a night-oriented schedule, the phase of PER1 and BMAL1 rhythms in PBMCs was delayed by ~2.5-3 h (P < 0.05), while no shift was observed for the other clock genes and the central markers. Three cycles of 8-h bright light induced significant phase delays (P < 0.05) of ~7-9 h for central and peripheral markers, except BMAL1 (advanced by +5h29; P < 0.05). Here, we demonstrate in humans a lack of peripheral clock adaptation under a night-oriented schedule and a rapid resetting effect of nocturnal bright light exposure on peripheral clocks.
