Adipose tissue stromal cells from lean, obese, and formerly obese mice

Project description:Adipose tissue stromal cells contribute to the regulation of adipose tissue in lean and obese states. Myeloid cells such as adipose tissue macrophages (ATMs) and dendritic cells (ATDCs) undergo both quantitative and qualitative changes with obesity. Due to similarity in markers the identify of adipose tissue dendritic cells and macrophages has been elusive. We have refined prior protocols to unambiguously discern ATM and ATDC in mice. We used microarrays to compare the profiles of ATMs and ATDC from gonadal adipose tissue from lean, obese, and formerly obese mice. We also isolated preadipocytes (PA) from lean and obese mice for comparison. Male C57Bl/6 mice were fed normal diet (ND) or high fat diet (HFD) for 16 weeks. Weight loss (WL) mice were switched from the HFD to ND for 8 weeks. RNA was purified from FACS sorted cell populations (live cells only) obtained from gonadal/epididymal adipose tissue depots. ATMs were defined as CD11c+ (CD45+CD64+ CD11c+) or CD11c- (CD45+CD64+ CD11c-) ATMs. ATDC were defined as CD64- CD11c+. PA were defined as CD31- CD45- Sca1+ PDGFRA+.

Subcutaneous adipose tissue from lean and obese subjects

Project description:Obtaining adipose tissue samples are paramount to the understanding of human obesity. We have examined the impact of needle-aspirated and surgical biopsy techniques on the study of subcutaneous adipose tissue (scAT) gene expression in both obese and lean subjects. Biopsy sampling methods have a significant impact on data interpretation and revealed that gene expression profiles derived from surgical tissue biopsies better capture the significant changes in molecular pathways associated with obesity. We hypothesize that this is because needle biopsies do not aspirate the fibrotic fraction of scAT; which subsequently results in an under-representation of the inflammatory and metabolic changes that coincide with obesity. This analysis revealed that the biopsy technique influences the gene expression underlying the biological themes commonly discussed in obesity (e.g. inflammation, extracellular matrix, metabolism, etc), and is therefore a caveat to consider when designing microarray experiments. These results have crucial implications for the clinical and physiopathological understanding of human obesity and therapeutic approaches. Expression data from bone marrow macrophage derived from lean and obese mice after 1,4, and 24h exposition to P.gingivalis

Project description:Obesity is a chronic inflammatory disease that weakens macrophage innate immune response to infections. Since M1 polarization is crucial during acute infectious diseases, we hypothesized that diet-induced obesity inhibits M1 polarization of macrophages in the response to bacterial infection. Using a computational approach in conjunction with microarray data, we identified switching genes that may differentially control the behavior of response pathways in macrophages from lean and obese mice. Bone marrow macrophages (BMMM-NM-&) from lean and obese mice were exposed to live Porphyromonas gingivalis (P. gingivalis) for three incubation times (1 h, 4 h and 24 h). cDNA from BMMM-NM-& of lean and obese mice were hybridized on Affymetrix Mouse Genome 430 2.0 Arrays. Hybridization was performed on three replicates at each time point (18 arrays total).

Project description:Macrophages are key to innate immunity and tissue homeostasis. Plasticity of macrophages allows rapid and highly orchestrated responses to various pathogenic and physiological challenges. Capturing the dynamic intracellular programs employed by macrophages during encounters with various stimuli presents a demanding technical challenge, and failures to fully characterize some of these responses has stalled development of disease-specific therapy. Powered by single-cell RNA sequencing (scRNA-seq), we expanded current t-distributed stochastic neighbor embedding (t-SNE) single-cell transcriptome algorithms and established a novel platform, MacSpectrum (macspec.github.io), with newly generated indexes (Macrophage Polarization Index, MPI and Macrophage Maturation Index, MMI) that allow not only macrophage subpopulation characterization under various conditions but also disease-specific signature gene set identification. Our MacSpectrum model will provide a comprehensive system to annotate macrophage function and dissect their diversified programs under sophisticated conditions in vivo, which represents a major challenge in macrophage biology. Single cell RNA sequeicing profiles of 6979 M0 BMDMs , 4736 M1 BMDMs, 6391 M2 BMDMs, 1710 lean ATMs, and 1758 obese ATMs were generated.

Project description:Adipose, once considered an inert storage depot, is now known to be an active endocrine tissue involved in total body homeostasis and metabolism, which exerts effects on multiple systems including food intake, immune function, and blood glucose regulation. Adipose tissue depots are known to have unique metabolic and gene expression profiles in vivo and when cultured in vitro. Differences in adipose tissue depot function could be important in determining chronic disease risk. Few comparisons of depot gene expression have been performed in the dog. Utilizing microarray technology, our objective was to identify differentially expressed genes and enriched functional pathways between subcutaneous and gonadal adipose of lean and obese dogs. Subcutaneous and gonadal adipose tissue samples were collected from 9 intact female beagles (4 yr-old; 4 lean controls; 5 obese ad libitum-fed) after 24 wk of ad libitum feeding.

Microarray of isolated adipocytes from subcutaneous adipose tissue of severely obese patients compared to lean controls

Project description:The aim of the project was to compare global gene expression in adipocytes from obese patients and lean controls. Subcutaneous adipose tissue was collected from severely obese patients undergoing bariatric surgery (average body-mass index (BMI) of 45.5 kg/m2 (n = 12, thereof 4 men) and healthy lean patients undergoing hernia repairs (average BMI of 24.2 kg/m2 (n = 12, thereof 7 men), between 27 and 56 years of age. Adipocytes were isolated by collagenase treatment of adipose tissue, followed by filtering and centrifugation. Floating adipocytes were lysed in Qiazol before RNA purification and microarray analysis.
