Project description:Hepatic molecular adaptations underlying periparturient metabolic diseases such as ketosis in dairy cows are largely unknown. We used a simple model for induction of ketosis to examine liver gene expression profiles using a microarray consisting of 13,257 annotated cattle oligonucleotides. At 4 days post-partum, 7 cows classified as healthy after a physical examination were fed at 50% of intake at day 4 from day 5 to signs of ketosis or until 14 days post-partum. Another group of 7 healthy cows served as controls. Liver was biopsied at 9-14 (ketosis) or 14 days post-partum (controls). Project description:This project is meant to document the effect of the negative energetic balance on the embryo quality and as it happens, to identify ways to improve the fertility of dairy bovines. Measurement of the β-Hydroxybutyric acid (BHB) has been taken between 45 and 60 days post-partum on twelve Holstein cows. Cows have been ranked according to the sanguine measurement as high or low in BHB in order to get 6 cows per group. After having synchronized the heat with hormones and an ovarian stimulation, cows have been inseminated and the embryos were selected as so to be transferred in at least two heifers. This project aim at isolating the differences created on the transcriptomic expression in morulas and blastocysts grown in cows with an energetic inbalance.

Project description:We aim at identifying possible differences in miRNA profiles and individual circulating miRNA between healthy cows and cows with persistent inflammation as diagnosed by cytology and histology. Ultimately the miRNA profiles will be correslated with RNAseq results to be performed from endometrial biopsies (planned later on this year following laser micro dissection). Samples have been taken from 6 "healthy cows" with low presence of immune cells at 21 and 44 days post partum which were subsequently pregnant, and from 11 cows with high numbers of immune cells at 21 and 44 days post partum, all non pregnant.

Project description:Heat stress (HS) has become a major challenge in the dairy industry around the world. Although numerous measures have been taken to alleviate the HS impact on milk production, the cellular level response to HS remains unclear in dairy cows. The objective of this study was to dissect functional alterations based on transcriptomic dynamics in the liver of cows under HS. Dairy cows exposed to HS exhibited both decreased feed intake and milk yield. Through liver transcriptomic analysis, differentially expressed genes were identified among three experimental conditions, including heat stress (HS), pair-fed (PF), and thermoneutral (TN) groups. We observed the upregulation of protein folding and inflammation-related genes in the HS group, while the mitochondrial genes were downregulated. Gene functional enrichment also revealed that mitochondria function and oxidative phosphorylation were dysregulated under HS. The liver transcriptome analysis generated a comprehensive gene expression regulation network upon HS in lactating dairy cows. Overall, this study provides novel insights into molecular and metabolic changes of cows conditioned under HS. Our results could facilitate the development of efficient biomarkers to mitigate the negative effect of HS on dairy cow health and productivity.

Project description:In this study, samples of 16 dairy cows from a MAP infected farm were used. Serum, milk and fecal samples were collected. Categorizing these cows into two groups based on their MAP infection status different standard methods for detection MAP were applied. Healthy controls showed no positive results in enzyme-linked immunosorbent assay (ELISA) with serum and milk samples (cattletype MAP Ab, Qiagen, Hilden, Germany; In-direct, IDVet, Grabels, France) and after cultivation of fecal samples on commercial Her-rold´s Egg Yolk Agars (HEYM agar, Becton Dickinson, Heidelberg, Germany) for 12 weeks. Cows with positive results were grouped into MAP infected cows. Specifically, for mass spectrometry analysis serum of seven MAP infected cows and seven healthy controls were used. All animals were from the same farm and were kept under the same environmental conditions. For additional mass spectrometry analysis with a further control group sam-ples of 21 dairy cows from an uninfected farm were examined. All cattle from this farm showed negative results in ELISA with serum and milk samples. Additionally, there was never a positive result in regularly tested fecal samples and sock swab samples of this farm. For verification of differential CTSS expression in Western blot analysis five dairy cows from another infected farm were consultedincluded. MAP status of these cows was analyzed by cultivation of fecal samples on HEYM agar for 12 weeks and ELISA with se-rum samples. In detail, two cattle were categorized into healthy controls and three cattle into MAP infected cows. Project description:In dairy cows, the transition from dry and pregnant to non-pregnant and lactating creates metabolic and immunological strain, which means many cows succumb to metabolic and infectious disease. This peripartum period, from three weeks pre-calving to three-weeks post-calving, is also known at the transition period. Metabolic health status during the transition period is determined by plasma concentrations of non-esterified fatty acids (NEFA), plasma β-hydroxybutyrate (BHBA), and liver triacylglycerol (TAG). High concentrations of these indicate cows that are under metabolic stress during the transition period and are, therefore, more likely to experience health conditions, also known as transition failure. Neutrophils, phagocytic cells of the innate immune response, are fundamental for fighting infectious agents.
