Project description:In F. graminearum, the transcriptional regulator TRI6 is encoded within the trichothecene gene cluster and regulates genes involved in the biosynthesis of the secondary metabolite deoxynivalenol (DON). Targeted disruption of TRI6 confirmed its role as a positive regulator of trichothecene genes and previous studies designated Tri6 as a pathway-specific transcriptional regulator. The Tri6 protein with its Cys2His2 zinc-finger may also conform to the class of broad-domain transcription regulators. This class of global transcriptional regulators mediate various environmental cues and generally responds to the demands of cellular metabolism. Expression profiling of F. graminearum grown under nitrogen-limiting conditions revealed that 49 out of 198 target genes are differentially regulated by TRI6. The identification of potential new targets together with deciphering novel binding site for Tri6, casts new light into the role of this transcriptional regulator in the overall growth and development of F. graminearum. Three biological replicates of Fusarium graminearum wildtype strain GZ3639 (NRRL 38155) (reference) and a tri6∆ mutant derived from GZ3639 were grown under nitrogen-limiting conditions in liquid culture for 5 hrs at 28oC

Project description:Fusarium fujikuroi is a biotechnologically important fungus due to its almost unique ability to produce gibberellic acids (GAs), a family of phytohormones. The fungus was described about 100 years ago as the causative agent of Bakanae (M-bM-^@M-^\foolish seedlingM-bM-^@M-^]) disease of rice. Apart from GAs, the fungus is known to produce pigments and mycotoxins, but the biosynthetic genes are known for only eight products. Here we present a high-quality genome sequence of the first member of the Gibberella fujikuroi species complex (GFC) that allowed de novo genome assembly with 12 scaffolds corresponding to the 12 chromosomes. In this work we focused on identification of all potential secondary metabolism-related gene clusters and their regulation in response to nitrogen availability by transcriptome, proteome, HPLC-FTMS and ChIP-seq analyses. We show that most of the cluster genes are regulated in a nitrogen-dependent manner, and that expression profiles fit to proteome and ChIP-seq data for some but not all clusters. Comparison with genomes of all available Fusarium species, including the recently sequenced F. mangiferae and F. circinatum, showed only a small number of common gene clusters and provides new insights into the divergence of secondary metabolism in the genus Fusarium. Phylogenetic analyses suggest that some gene clusters were acquired by horizontal gene transfer, while others were present in ancient Fusarim species and have evolved differently by gene duplications and losses. One polyketide synthase (PKS) and one non-ribosomal peptide synthetase (NRPS) gene cluster are unique for F. fujikuroi. Their products were identified by combining overexpression of cluster genes with HPLC-FTMS-based analyses. In planta expression studies suggest a specific role of the PKS19 product in rice infection. Our results indicate that comparative genomics together with the used genome-wide experimental approaches is a powerful tool to uncover new secondary metabolites and to understand their regulation at the transcriptional, translational and epigenetic levels. Project description:MicroRNAs (miRNAs) are a class of endogenous non-coding small RNAs that regulate targeted mRNAs by degrading or repressing translation, considered as post-transcrption regulators. So far, a large number of miRNAs have been discovered in model plants, but little information is available on miRNAs in banana. In this study, by sequencing the small RNA (sRNA) transcriptomes of Fusarium wilt resistant and susceptible banana varieties, 139 members in 38 miRNA families were discovered, and six out of eight new miRNAs were confirmed by RT-PCR. According to the analysis of sRNA transcriptome data and qRT-PCR verification, some miRNAs were differentially expressed between Fusarium wilt resistant and susceptible banana varieties. Two hundred and ninety-nine and 31 target genes were predicted based on the draft maps of banana B genome and Fusarium oxysporum (FOC1, FOC4) genomes respectively. Specifically, two important pathogenic genes in Fusarium oxysporum genomes, feruloyl esterase gene and proline iminopeptidase gene, were targeted by banana miRNAs. These novel findings may provide a new strategy for the prevention and control of Fusarium wilt in banana.

Project description:Resistance mechanisms of wheat to Fusarium graminearum are still poorly understood. Multiple reports have investigated transcriptomic differences between resistant and susceptible genotypes. To investigate the genetic determinants underlying the two major contributor QTL Fhb1 and Qfhs.ifa-5A, we generated a set of near-isogenic lines, that we have used for transcriptomic studies using the Affymetrix wheat GeneChip®: 4 near-isogenic lines (BC5F2) derived from a cross of CM82036 and cv Remus (susceptible recurrent parent) and the resistant parent CM82036 have been inoculated with Fusarium spore suspension or water as control at anthesis. Samples have been taken at 3 timepoints 8, 24 and 72 hours after inoculation. Five wheat heads per condition were pooled into one sample, that has been used for RNA preparation. Including 3 replicates a total of 90 microarrays have been hybridized. Our results identify a wide variety of genes (631 transcripts) that are differentially regulated for Fusarium graminearum in all genotypes after 72 h and 339 genes that are upregulated only in lines without Qfhs.ifa.5A. Few genes were found differentially regulated for Fusarium and QTL. Project description:Crown rot of wheat, caused by Fusarium pseudograminearum and other Fusarium species is an important disease globally. To understand the host response to challenge by Fp, we examined gene exression changes in the stem base of the wheat variety Kennedy, following inoculation with macroconidia using the Affymetrix GeneChip Wheat Genome Array. Induced genes included mainly those with defensive functions such as genes encoding anti-microbial proteins as well as oxidative stress-related proteins, signalling molecules, and proteins involved in both primary and secondary metabolism. This study is the first comprehensive analysis of the wheat transcriptome during crown rot infection and provides new insights into the host processes involved in plant defence against this pathogen. Experiment Overall Design: There are six samples, three F. pseudograminearum inoculated samples and three mock inoculated samples. Each sample consists of 2cm of stem base from approximately 20 plants.

Project description:Fusarium graminearum (teleomorph Gibberella zeae) is a prominent pathogen that infects major cereal crops, such as wheat, barley, and maize. Conidiogenesis had been intensively studied in Aspergillus nidulans and regulatory pathway genes have been known to regulate conidiogenesis in stage specific manner. We reported the functional analyses of flbD, abaA, and wetA orthologs in F. graminearum. To understand genome-wide transcriptional profiling of conidiation, we employed RNA-seq of the wild-type Fusarium graminearum Z-3639 and each gene deletion mutants with three time courses (0 h, 6 h and 12 h after induction of conidiogenesis). Project description:Fusarium oxysporum causes Fusarium wilt syndrome in more than 120 different plant hosts, including globally important crops such as tomato, cotton, banana, melon, etc. F. oxysporum shows high host specificity in over 150 formae speciales and have been ranked in the top 10 plant fungal pathogens. Although three PMTs encoded by the pmt1, pmt2, and pmt4 are annotated in the genome of F. oxysporum, their functions have not been reported. As O-mannosylation is not found in plants, a comprehensive understanding of PMTs in F. oxysporum becomes attractive for the development of new strategy against Fusarium wilt. In order to understand the molecular mechanism of the differential functions of three PMTs, a comparative O-glycoproteome analysis of the pmt mutants were carried out.
