Project description:Pluripotent stem cells fluctuate among distinct functional cell states. Such heterogeneity undermines the potential applications of stem cells in regenerative medicine. The heterogeneity of pluripotent cell states and their molecular nature remain less understood. Here, we utilized mass cytometry to dissect pluripotent cell states by simultaneously profiling multiple proteins and their modifications in single cells. Results indicated the role of extensive signaling cross-talk among multiple pathways in regulating cell state heterogeneity. By systematically dissecting the signals and their combinations, we identify conditions through which large portions of pluripotent cells can be chemically reprogrammed into totipotent cells which can be cultured. Unlike pluripotent cells, these chemically induced totipotent cells (CiTC) give rise to both embryonic and extra-embryonic cell types. In addition, CiTCs are molecularly distinct from pluripotent cells. CiTCs provide an in-vitro model to understand the nature of totipotent cells and the plasticity of pluripotent cells. Our quantitative and integrative approach provides general method to understand similar heterogenous cell systems such as cancer.

Project description:We demonstrate induction and long-term maintenance of totipotent stem cells (TotiSCs) from mouse pluripotent stem cells (PSCs) by a combination of three small molecules, TTNPB, 1-Azakenpaullone, and WS6. These cells, which we designated as ciTotiSCs (chemically induced totipotent stem cells), resembled mouse totipotent 2C-embryo stage cells at both transcriptome and epigenome level.

Multi-parametric MRI for Therapeutic Response Prediction in Rectal Cancer

Project description:Interventions: Single arm prospective observationaI imaging biomarker study. Patients will have standard treatment, consisting of neoadjuvant chemoradiotherapy followed by surgery. There will be no change to the patient’s treatment by participating in this study. Patients participating in this study will have multi-parametric MRI and PET/CT at the following 3 time-points: 1. Prior to chemoradiotherapy 2. During the third week of chemoradiotherapy 3. Post chemoradiotherapy, within 1 week prior to surgery. A multi-parametric MRI incorporates standard morphological as well as diffusion weighted imaging and dynamic contrast enhanced sequences. The total follow-up duration will be 2 years from date of surgery. Primary outcome(s): Correlation of multi-parametric MRI (diffusion weighted imaging (DWI) and dynamic contrast enhanced (DCE)) at 3 Tesla with surgical histopathology tumour regression grade (TRG). Histopathological assessment will be performed on surgical specimens. TRG assessment will be performed as per the modified classification of Ryan et al set out in the AJCC Cancer Staging Manual, 7th Edition as follows: TRG 0 (complete response) - no viable cancer cells TRG 1 (moderate response) - single cells or small groups of cancer cells TRG 2 (minimal response) - residual cancer outgrown by fibrosis TRG 3 (poor response) - minimal or no tumour kill; extensive residual cancer. [At baseline pre-chemoradiotherapy, week 3 of chemoradiotherapy, and post-chemoradiotherapy (within 1 week prior to surgery) ]
