Project description:Adult T-cell leukemia/lymphoma (ATLL) patients are in severely poor prognosis partially because of their immunosuppressive state. As majority of patients have leukemic cells with the similar phenotype with Tregs, some reports have suggested that ATLL cells themselves have immunosuppressive functions. However, hitherto, there are controvertial discussions. In this study, we found that CD39 is expressed on some of ATLL cells, especially highly on those of aggressive subtypes.

Project description:Adult T-cell leukemia/lymphoma (ATLL) patients have an extremely poor prognosis, partly due to their immunosuppressive state. The majority of ATLL patients have leukemic cells with phenotype similar to Tregs, prompting suggestions that ATLL cells themselves have immunosuppressive functions. In this study, we detected CD39 expression on ATLL cells, particularly frequent on aggressive subtypes. CD39 and CD73 convert extracellular adenosine triphosphate (ATP) into adenosine, a key player in Tregs' immunosuppression. In vitro culture, both CD39+ ATLL cells and normal Tregs converted rapidly extracellular ATP to AMP, which was disturbed by CD39 inhibitors, and was negated in the CD39 knockout MJ cell line. The proliferation of cocultured CD4+/CD8+ normal T cells was suppressed by CD39+ MJ cells, but not by CD39 knockout MJ cells. Supplemented ATP was exhausted by an EG7-OVA T-cell line with stable CD39 induction, but not by mock. When these cell lines were subcutaneously transplanted into murine flanks, Poly(I:C) peritoneal administration reduced tumor size to 1/3 in mock-transplanted tumors, but not in CD39 induced tumors. Overall, we found that ATLL cells express CD39 at a high rate, and our results suggest that this helps ATLL cells escape antitumor immunity through the extracellular ATPDase-Adenosine cascade. These findings will guide future clinical strategies for ATLL treatment.

Microglia regulate basal cerebral blood flow and neurovascular coupling through ectonucleotidase CD39 II

Project description:Microglia modulate cerebral blood flow and cerebrovascular reactivity, but the mechanisms remain elusive. Here we show that pharmacological or genetic method induced microglia-ablation, which causes reduction of the microglia-expressed ATP/ADP-hydrolyzing ectonucleotidase CD39, elevates the cerebral blood flow baseline, while reducing the cerebral blood flow change to whisker stimulation and intra-cisterna magna injection of ATP, but not adenosine. Microglia P2ry12 inhibition with clopidogrel increased basal cerebral blood flow and the effect was abolished in microglia specific CD39 knockout mice. P2ry12 knockout mice show reduced cerebral blood flow changes to whisker stimulation or intra-cisterna magna injection of ATP. Microglia-specific deletion of CD39 or its pharmacological inhibition reproduces the effects of microglia ablation on cerebral blood flow and attenuates whisker stimulation-induced adenosine concentration increase in contralateral barrel cortex. These results suggest that microglia-dependent adenosine production is an integral component of the brain purinergic signaling system that governs neurovascular coupling.

Project description:Microglia tightly interact with brain capillaries and regulate blood brain barrel permeability in pathological conditions. However, it is still unknown whether and how microglia regulate CBF in homeostatic condition. Here we show microglia directly regulate cerebral blood flow (CBF) and neurovascular coupling. Conditional knockout of microglial CD39 resulted in CBF hyper perfusion, reduced ATP induced CBF increase, impaired neurovascular coupling by whisker stimulation and reduced whisker stimulation induced adenosine increase in barrel cortex. Out data suggest that microglia is an important player for CBF regulation.

Comparison of human CD39- and CD39+ thymus-derived regulatory T-cells

Project description:CD39- and CD39+ human thymus-derived regulatory T-cells (tTreg) present as distinct subsets with specific functional and metabolic response patterns. Both cell types express equal levels of the canonical tTreg transcription factors FOXP3 and Helios and overexpression studies have shown that CD39 expression is independent of these two transcription factors. So far, no transcriptomic analyses to adress these differences have been performed. We used microarrays to detail the global programme of gene expression between CD39- and CD39+ human tTreg and identified distinct classes of up-regulated and down-regulated genes between these two subsets.

Distinctive CD39+CD9+ lung interstitial macrophages suppress IL-23/Th17-mediated neutrophilic asthma by inhibiting NETosis [scRNA-Seq]

Project description:IL-23-Th17 signaling axis is responsible for neutrophilic inflammation in various barrier tissues. However, mechanistic links between IL-23 and Th17 activation remain unclear, despite of their critical contribution to chronic inflammatory diseases. Here, we discovered a novel signaling pathway linking IL-23 to Th17 activation in our neutrophil-dominant asthma (NDA) model. Through single-cell multi-omics analysis, we identified distinctive CD39+CD9+ interstitial macrophages (IMs) suppressed by IL-23. CD39+CD9+ IMs increased by IL-23p19 inhibitor suppressed NETosis, which inhibited Th17 activation. To inhibit NETosis, CD39+CD9+ IMs first attach to neutrophils in a CD9-dependent manner, and then remove ATP molecules near neutrophils in a CD39-dependent manner. Thus, our discovery of CD39+CD9+ IMs provides previously unrecognized signaling component linking IL-23 and NETosis for controlling Th17 activation/neutrophilic inflammation. We finally demonstrated the decreased number of CD39+CD9+ immune cells in patients with severe chronic rhinosinusitis and inflammatory bowel disease, suggesting CD39+CD9+ IMs as potential therapeutic targets for IL-23-Th17-mediated inflammatory diseases.
