Project description:The search of the molecular process associated with development and metabolism of muscle mass is still actual, what in humans is related to muscle modification after endurance training exercises. In turn in farm animals, the high meat content in carcasses delivers higher economic benefit. Over a few recent years, scientists using next-generation-sequencing (NGS) methods attempted to indicate proteins/genes engaged in muscle mass development. The aim of the present study was to show mRNA degradome signals that could be characteristic for molecular processes associated with muscle development in pigs and indicate the miRNA regulation controlling this process based using also NGS technology. The preliminary results exhibited that the huge amount of miRNAs regulate gene expression at the different molecular level than mRNA degradation. Moreover, the study found significant differences between investigated pig breeds. The results often were opposite for Pietrain breed that could be related to differences in muscle fibre composition and their microstructure. The present study showed an interesting dependence between miRNA and their targets - the most of transcripts found as being degraded, were associated with controlling muscle mass development such as ENO3, CKM, CRYAB and ADAM19. In turn, performed PAREsnip analysis showed that only a small percentage of identified degraded transcripts was associated with miRNA regulation, what suggested a different mechanism of mRNA degradation. Nevertheless, the complete view of miRNA regulation could be obtained in the more advanced study, which would employ miRNA transfection procedure in muscle tissue cell culture that could be a subject of further research.

Project description:Loss of muscle mass is a pathological driver in both chronic illness and aging. We have discovered that skeletal muscle specific thrombospondin-1 (Thbs1) transgenic mice have profound muscle atrophy with age-dependent decreases in exercise capacity and premature lethality. Here, we compared the transcriptional profiles of skeletal muscle from control non-transgenic mice with those overexpressing Thbs1 in order to elucidate the transcriptional changes underlying the observed effects on skeletal muscle mass. The overall purpose was to identify new, effective targets to combat muscle atrophy and sarcopenia.

FBXW7 coordinates postprandial muscle glucose metabolism and maintenance of skeletal muscle mass

Project description:In this study, we show that muscle-specific inactivation of FBXW7 elicits striking defects in postprandial glucose metabolism and failure to maintain skeletal muscle mass in adult mice. Further, mice lacking FBXW7 exhibited impaired endurance capacity and exacerbated HFD-induced insulin resistance and postprandial hyperglycemia. At the mechanistic level, RNAseq and quantitative proteomic analysis revealed global effects of FBXW7 deficiency on skeletal muscle transcriptome and proteome. This work illustrates a prominent role of FBXW7 in integrating postprandial nutritional signals to coordinate glucose metabolism and muscle mass maintenance.

Smad2 and 3 transcription factors control muscle mass in adulthood

Project description:Loss of muscle mass occurs in a variety of diseases including cancer, chronic heart failure, AIDS, diabetes and renal failure, often aggravating pathological progression. Preventing muscle wasting by promoting muscle growth has been proposed as a possible therapeutic approach. Myostatin is an important negative modulator of muscle growth during myogenesis and myostatin inhibitors are attractive drug targets. However, the role of the myostatin pathway in adulthood and the transcription factors involved in the signaling are unclear. Moreover recent results confirm that other TGFβ members control muscle mass. Using genetic tools we perturbed this pathway in adult myofibers, in vivo, to characterize the downstream targets and their ability to control muscle mass. Smad2 and Smad3 are the transcription factors downstream of myostatin/TGFβ and induce an atrophy program which is MuRF1 independent and requires FoxO activity. Furthermore Smad2/3 inhibition promotes muscle hypertrophy independent of satellite cells but partially dependent of mTOR signalling. Thus myostatin and Akt pathways cross-talk at different levels. These findings point to myostatin inhibitors as good drugs to promote muscle growth during rehabilitation especially when they are combined with IGF1-Akt activators.

Estrogen receptor β regulates skeletal muscle mass and muscle stem cell proliferation in young female mice

Project description:The female sex hormone estrogens plays a critical role in maintaining muscle mass and muscle stem cell (MuSCs) functions. However, it is still unclear about downstream pathways of estrogens including its receptors that are expressed in both skeletal muscle tissue and MuSCs. To study the specific role of estrogen receptor β (ERβ), one of two main types of estrogen receptors, in skeletal muscle and MuSCs, we generated muscle-specific ERβ-knockout (mKO) mice and muscle stem cell-specific ERβ-knockout (scKO) mice. Here, we show that muscle-specific ERβ-deficient induced decreased muscle strength and fast-type muscle mass in young female mice. Furthermore, muscle stem cell-specific ERβ-deficient young female mice but not male exhibited impaired muscle regeneration ability after acute muscle injury, accompanied by a decreased proliferation rate of muscle stem cells. RNA sequencing analysis showed that the loss of ERβ in muscle stem cells changes the expression of cell cycle associated genes and niche component factors including laminin and collagen. Thus, our characterization of mKO and scKO mice indicate that the estrogen-ERβ pathway is a sex-specific regulatory mechanism that controls both skeletal muscle mass and the proliferation of muscle stem cell in females and could be of importance in a therapeutic context.
