Project description:The plasma protein FHR1 induces release of inflammatory cytokines IL-1β, IL-6, IL-18 or TNFα from blood-derived human monocytes. RNA sequencing was performed from RNA of BSA- or FHR1-treated monocytes from 4 different donors. In response to FHR1, 522 monocytic genes were upregulated (gene ontology enrichment analysis), including 35 inflammation related genes, e.g. TNF. Also, G protein-coupled receptors such as EMR2/ADGRE2 were upregulated in response to FHR1.

Early RNA sequencing of human monocytes in presence of Candida albicans

Project description:C.albicans induces the upregulation of inflammation related genes at the same time it also induces TGF-ß signalling pathway related genes from human blood derived monocytes. RNA sequencing was prerformed from Candida albicans co-incubated monoyctes from 3 different donors. Candida albicans significantly upregulates 6363 genes in human blood derived monocytes in 1h of co-incubation.

RNA-sequencing analysis of effect of miR-155 overexpression in human CD14+ monocytes

Project description:We report the application of bulk RNA-Sequencing to assess the effect of miR-155 overexpression by profiling gene expression following transfection of miR-155 mimic compared against negative control mimic in human peripheral blood CD14+ monocytes. Isolated human CD14+ monocytes were transfected for 24 hours and mRNA profiles of miR-155 and negative control mimic were generated by bulk RNA-Sequencing in 5 donors. The sequence reads passed quality checks and were analysed using DeSeq2. We mapped the sequence reads to the human genome (build hg38) and identified 16,014 transcripts.

Long non-coding RNA landscape in human monocytes

Project description:We report the lncRNAs transcribed in the human primary monocyte cells. We performed deep RNA sequencing from four healthy individuals. In addition, the raw RNA-Seq data from 11 human monocyte samples were selected from public databases and generated a total ~1.7 billion reads. We identified ~ 8,000 lncRNAs from all the datasets of which more than 1,000 of them have not been previously reported in monocytes. We also validated a few of these novel lncRNAs in monocytes and other hematopoetic cell types. Staphylococcus aureus Mediated Tolerance Induction in Human Monocytes is accompanied with Specific Alterations in Membrane Receptor Expression

Project description:Exposure of human monocytes to lipopolysaccharide (LPS) or other pathogen-associated molecular pattern (PAMPs) induces a temporary insensitivity to subsequent LPS challenges, a cellular state called endotoxin tolerance (ET). The tolerant state of monocytes is accompanied by cell surface and other glycoprotein expression changes induced by the activation of Toll-like receptors (TLRs). In this study, we aimed to characterize the cellular state of human monocytes stimulated with Gram-positive Staphylococcus aureus and TLR2 ligands. We analyzed gene expression changes induced by S. aureus after 2 and 24 hours by amplicon sequencing (RNA-AmpliSeq) and compared the pro-inflammatory response after 2 hours of stimulation to the to the response in re-stimulation experiments after the first stimulus. In parallel, glycoprotein expression changes in human monocytes after 24 hours of S. aureus stimulation were analyzed by proteomics and compared to stimulation experiments with TLR2 ligands Malp-2 and Pam3Cys and TLR4 ligand LPS. The results demonstrate that monocytes stimulated with S. aureus and TLR ligands entered the tolerant cell state after activation. Compared to TLR agonist mediated activation and tolerization of monocytes, glycoprotein expression changes induced by S. aureus stimulation revealed significant differences in receptor expression profiles. We report a glycoprotein expression profile characteristic for PAMP and S. aureus tolerized human monocytes. Finally, we analyzed peripheral blood monocytes of patients with S. aureus bloodstream infection for inflammatory responses in vitro and for their glycoprotein expression profiles. RNA-AmpliSeq data from patient-derived monocytes demonstrated that the cells were pro-inflammatory responsive to S. aureus stimulation and expressed higher level of CD44 mRNA, while other markers of the tolerant cell state were not detected.
