Project description:Various studies showed a significant production/release of IL-1 beta following status epilepticus (SE) and during epileptogenesis. We studied the role of this cytokine in the epileptogenic process by blocking its production and downstream effects in vivo in a model of acquired epilepsy following SE induction. We used a pharmacological strategy consisting of the administration of two drugs, namely VX-765 and Anakinra, which selectively block the biosynthesis and the receptor type 1 of this cytokine, respectively.

Project description:The study of HIV infection and pathogenicity in physical reservoirs requires a biologically relevant model which resembles the human immune system and physiology. The human immune system (HIS) mouse is an established model of HIV infection1–3, but defects in immune tissue reconstitution remain a challenge for examining pathology in tissues1,3–7. Herein we show that exogenous injection of the human cytokine FLT-3L into the hematopoietic stem cell (HSC) cord blood HIS mouse model significantly expanded the total area of axillary lymph nodes and the absolute number of circulating human T cells, thus enabling us to visualize and quantify HIV infectivity in the secondary lymphoid tissues of the spleen and axillary node. Further, we detected cell death and human T cell depletion in tissues, consistent with HIV pathogenesis. Treatment with the Caspase-1 inhibitor VX-765 restored CD4+ T donor cells and decreased plasma viral load as measured by gag qPCR, and apoptosis in the spleen. In situ hybridization further demonstrated a decrease in viral RNA in both the spleen and axillary lymph nodes. Transcriptomic analysis revealed that in vivo inhibition of caspase-1 led to an upregulation in host HIV restriction factors including APOBR, SAMHD1 and APOBEC3A (study design depicted in graphical abstract). These findings demonstrate that exogenous rFLT-3L as a mechanism to enhance human immune system characteristics in HIS mice. These enhancements will support investigations of HIV pathogenesis and immune outcomes in tissue compartments. Targeting inflammasome pathways with VX-765 in HIS mice treated with rFLT-3L preserved T cell populations and decreased viral load after HIV infection.

Project description:Purpose: To assess if NLK modulates ER target gene expression, we performed transcriptome sequencing following NLK inhibition or tamoxifen treatment in the BT483 or T47D-TamR cells under estrogen-deprived condition. Methods: BT483 and T47D-TamR cells were treated with NLK-specific siRNAs, or VX-702, or Tamoxifen under estrogen-deprived condition to examine the expression profile changes of ER target genes. NovaSeq 6000 S2 Reagent Kit (Illumina) was used for paired-end reads (2×150 bp) sequencing reactions. The expression changes of ER target genes following NLK inhibition are compared to their changes following tamoxifen treatment. Results: Our results show that the ER target gene expression changes following NLK silencing or VX-702 treatment correlate with their expression changes following tamoxifen treatment.

Project description:Organ-on-a-chip platforms are utilized in global bioanalytical and toxicological studies as a way to reduce materials and increase throughput as compared to in vivo based experiments. These platforms bridge the infrastructure and regulatory gaps between in vivo animal work and human systems, with models that exemplify active biological pathways. In conjunction with the advent of increased capabilities associated with next generation sequencing and mass spectrometry based ‘-omic’ technologies, organ-on-a-chip platforms provide an excellent opportunity to investigate the global changes at multiple biological levels, including the transcriptome, proteome and metabolome. When investigated concurrently, a complete profile of cellular and regulatory perturbations can be characterized following treatment with specific agonists. In this study, global effects were observed and analyzed following liver chip exposure to the chemical warfare agent, VX. Even though the primary mechanism of action of VX (i.e. acetylcholinesterase inhibition) is well characterized, recent in vivo studies suggest additional protein binding partners that are implicated in metabolism and cellular energetic pathways. Also, secondary toxicity associated with peripheral organ systems, especially in human tissues, is not well defined. Our results demonstrate the efficacy of utilizing an organ-on-a-chip platform as a surrogate system to traditional in vivo studies. This is realized by specifically indicating significant dysregulation of a number of cellular processes in response to VX exposure including but not limited to amino acid synthesis, drug metabolism, and energetics pathways.

Project description:We performed mRNA sequencing of reciprocal F1 female hybrids from two crosses (362/765 and 517/765) and the parental DGRP lines (362, 517 and 765). Specifically, we aimed to identify whether transcripts predicted to be regulated by cis-eQTLs exhibit a significant allele-specific bias in gene expression. Since both alleles act in the cross in the same trans environment, differential expression in the F1 is a direct measure of cis-regulatory activity.

Project description:We used small RNA seq (Illumina) to characterise and quantify the miRNA and small non-coding (snc)RNA transcriptome of 38 libraries of size-fractionated RNA obtained from AML patients and normal bone marrow. We detected the expression of 765 known miRNAs and 2554 uniquely located species of sncRNAs. A total of 575 potential miRNA candidate hairpins were identified. We also identified 37 C/D Box and 51 H/ACA Box, potential novel snoRNAs.
