Project description:Bariatric surgical techniques are known to cause weight loss and diabetes remission to varying degrees in severly obese patients. However, the mechanisms involved in the restoration of beta-cell function remain to be uncovered. In this study, the leptin-deficient ob/ob mouse was used as a model to investigate the effect of EGA bariactric surgery on pancreatic islet miRNA expression.

Project description:Ob/ob mice are characterized by a defect in leptin synthesis. In this study, the contribution of leptin deficiency to the deregulation of miRNA expression in ob/ob mice was determined by comparing leptin infused ob/ob mice to saline control.
Project description:Purpose:To identify which genes are differentially expressed in pancreatic islets of the ob/ob mice compared to the control ob/+ mice Methods: The data were aligned to mm10 reference genome with HISAT2 (V2.1.0).Differential expression analysis was performed using the DESeq2(V 1.2.0). Results:Library construction, and sequencing were performed by Beijing Genomic Institution (www.genomics.org.cn, BGI) using an optimized data analysis workflow
Project description:GWAT store most of the TAG in mice, ob/ob mice is an obese mice. Ob/ob/Fsp27-/- mice are lean when compared with ob/ob mice. The GWAT weight was dramatically reduced in ob/ob/Fsp27-/- mice. We next extract the total RNA from the GWAT of ob/ob and ob/ob/Fsp27-/- mice, to perform microaary analysis using Mouse Genome 430 2.0 arrays, Affymetrix. We then analysised the up-regulated and down regulated pathways.
