Carbon and Nitrogen Isotope Ratios of Food and Beverage in Brazil.

Project description:Several previous studies on targeted food items using carbon and nitrogen stable isotope ratios in Brazil have revealed that many of the items investigated are adulterated; mislabeled or even fraud. Here, we present the first Brazilian isotopic baseline assessment that can be used not only in future forensic cases involving food authenticity, but also in human forensic anthropology studies. The δ13C and δ15N were determined in 1245 food items and 374 beverages; most of them made in Brazil. The average δ13C and δ15N of C3 plants were -26.7 ± 1.5‱, and 3.9 ± 3.9‱, respectively, while the average δ13C and δ15N of C4 plants were -11.5 ± 0.8‱ and 4.6 ± 2.6‱, respectively. The δ13C and δ15N of plant-based processed foods were -21.8 ± 4.8‱ and 3.9 ± 2.7‱, respectively. The average δ13C and δ15N of meat, including beef, poultry, pork and lamb were -16.6 ± 4.7‱, and 5.2 ± 2.6‱, respectively, while the δ13C and δ15N of animal-based processed foods were -17.9 ± 3.3‱ and 3.3 ± 3.5‱, respectively. The average δ13C of beverages, including beer and wine was -22.5 ± 3.1‱. We verified that C-C4 constitutes a large proportion of fresh meat, dairy products, as well as animal and plant-based processed foods. The reasons behind this high proportion will be addressed in this study.

Biological nitrogen removal from low carbon wastewater

Project description:Nitrogen has traditionally been removed from wastewater by nitrification and denitrification processes, in which organic carbon has been used as an electron donor during denitrification. However, some wastewaters contain low concentrations of organic carbon, which may require external organic carbon supply, increasing treatment costs. As a result, processes such as partial nitrification/anammox (anaerobic ammonium oxidation) (PN/A), autotrophic denitrification, nitritation-denitritation and bioelectrochemical processes have been studied as possible alternatives, and are thus evaluated in this study based on process kinetics, applicability at large-scale and process configuration. Oxygen demand for nitritation-denitritation and PN/A is 25% and 60% lower than for nitrification/denitrification, respectively. In addition, PN/A process does not require organic carbon supply, while its supply for nitritation-denitritation is 40% less than for nitrification/denitrification. Both PN/A and nitritation-denitritation produce less sludge compared to nitrification/denitrification, which saves on sludge handling costs. Similarly, autotrophic denitrification generates less sludge compared to heterotrophic denitrification and could save on sludge handling costs. However, autotrophic denitrification driven by metallic ions, elemental sulfur (S) and its compounds could generate harmful chemicals. On the other hand, hydrogenotrophic denitrification can remove nitrogen completely without generation of harmful chemicals, but requires specialized equipment for generation and handling of hydrogen gas (H2), which complicates process configuration. Bioelectrochemical processes are limited by low kinetics and complicated process configuration. In sum, anammox-mediated processes represent the best alternative to nitrification/denitrification for nitrogen removal in low- and high-strength wastewaters.

Anaerobic carbon-limited retentostat cultivation of Lactobacillus plantarum WCFS1

Project description:Lactobacillus plantarum WCFS1 was grown under anaerobic carbon-limited conditions in a chemostat with complete biomass retention (retentostat). In this cultivation system, the biomass concentration progressively increases while the dilution rate is kept constant, resulting in decreased specific susbtrate availibility, and hence, a progressive decrease in the specific growth rate. During the progressive transition from growth to virtually no growth, the global changes occurring at the level of metabolism and gene expression were studied using a genome-scale metabolic model and DNA microarrays. Four different time-points are compared, corresponding to 4 different specific growth rates, and hence, 4 different ratios of energy used for maintenance and growth. The samples taken at the start of retentostat cultivation serves as a a reference sample, to which the three other samples (taken after 3 days, 17 days, and 31 days under retentostat conditions) are compared. No biological replicates: all samples were taken from the same retentostat fermentation.
