Project description:Clinical observations suggest increased size of lens in highly myopic subjects compared with emmetropic subjects. The layer of anterior lens epithelial cell membrane, being the most metabolically active part of the lens, can be easily acquired during standard lens surgery and is an ideal sample to study the gene expression difference between lens of different populations. In this study, to investigate the possible molecular mechanisms underlying different features of lens between highly myopic and emmetropic subjects, gene expression profiling of lens epithelia of corresponding population was conducted.

microRNA profiling in the aqueous humor of highly myopic eyes by next generation sequencing

Project description:Purpose: To characterize microRNAs (miRNAs) and their possible roles in high myopia by using next generation sequencing Methods: Aqueous humor samples were obtained from 15 highly myopic eyes and 15 cataract eyes at the onset of surgery. miRNA next generation sequencing and bioinformatics analyses were performed using RNA extracted from aqueous humor samples. Results: A total of 341 miRNAs were detected in the aqueous humor samples of highly myopic eyes; 201 miRNAs were detected in the aqueous humor samples of cataractous control eyes. A total of 249 mature miRNAs and 17 novel miRNAs were differentially expressed during myopia. Possible pathways regulated by these aberrantly expressed miRNAs included the TNF, MAPK, PI3K-Akt, and HIF-1 signaling pathways. Conclusions: The current study provided an overall view of miRNA profiling in the aqueous humor of highly myopic eyes. These profiles may be associated with myopia pathogenesis, and are potential biomarkers.

Project description:The pathogenesis of myopia involves different molecular mechanisms and pathways, which can be reflected in the aqueous humor. In our study, aqueous humor was collected for liquid chromatograph mass spectrometer (LC/MS) analysis from 30 individual eyes that underwent phacoemulsification and intraocular lens (IOL) implantation. A total of 190 DEPs (differentially expressed proteins) were identified, which revealed their involvement in pathways such as carbon metabolism, lipid, and cholesterol metabolism, ECM (extracellular matrix) component regulation, and scleral remodeling in the context of high myopia. Further analysis revealed 15 interacting hub proteins, encompassing functions related to lipoprotein assembly, regulation, complement cascades, and fibrin formation. Subsequent validation demonstrated elevated levels of APOE (apolipoprotein E), C3 (complement 3), and AHSG (alpha-2-HS-glycoprotein) in the high myopia group (31 eyes of cataracts and 45 eyes of high myopia with cataracts), while AHSG had a significant positive correlation with axial length in patients with high myopia and good efficacy in distinguishing between myopic and non-myopic groups. These results suggested that the interaction of lipoproteins, complement cascades, and the fibrinolytic system played a pivotal role in the pathological process of myopia, while the metabolism of essential materials and the remodeling of ECM and tissue structure were affected in the eyes of individuals with high myopia.

Tissue specific comprehensive gene analysis in a murine lens-induced myopia model

Project description:To identify to identify target tissues and molecules involved with refractive myopic shift and axial length elongation in a murine lens-induced myopia model, we performed comprehensive analysis by microRNA array. Negative 30diptor (-30D) lens was fixed on right eye (-30D) of C57BL/6J mice (3weeks old, N=3) for 3 weeks, the refraction and the axial length were measured using a refractometer and a SD-OCT system in all eyes. Eye balls were enucleated and separated to cornea, iris, lens, retina, choroid and sclera. Total RNA was extracted from individual ocular components. MicroRNA expression analysis was carried out using Agilent Mouse miRNA Microarray (8×60K) miRBase21.0 (Agilent). Expression ratio calculation and miRNA varying expression were extracted by GeneSpring GX 14.5 (Agilent). After 3weeks of lens fixing, a refractive change and an axial length elongation change were observed (Normal vs -30D: 0.95 ± 1.85D vs -18.42 ± 3.98D, 0.155 mm ± 0.015mm vs 0.273 ± 0.009 mm), respectively. MiRNA expression changes that induced only by -30D lens fixing was confirmed in each part of the eyeball. By expression ratio calculation and miRNA varying expression analysis, upregulated miRNA (56 in cornea, 13 in iris, 6 in lens, 0 in retina, 29 in choroid and 30 in sclera) and downregulated miRNA (7 in cornea, 28 in iris, 17 in lens, 9 in retina, 7 in choroid and 40 in sclera) were observed. Overlapping miRNAs were also found while each eye tissues. In this study, miRNA varying expression were observed in each ocular part of the murine lens-induced myopia model. These miRNAs dysregulation may be functionally involved with the refractive myopia shift and the axial length elongation.

Transcription profiling of chicken retinal gene expression during imposed myopic defocus

Project description:The retina plays an important regulatory role in ocular growth. To screen for new retinal candidate genes that could be involved in the inhibition of ocular growth, we used chick microarrays to analyze the changes in retinal mRNA expression after myopic defocus was imposed by positive lens-wear. Chicks were raised under a 12h-light/12h-dark cycle (light-onset: 8:00 am and light-offset: 8:00 pm) with unrestricted access to water and food. On the day prior to the experiment, velcro rings were glued to the feathers around the eye under diethylether anaesthesia. Four male white leghorn chicks, aged 9 days, wore +6.9D spectacle lenses over both eyes for 24 hours. Four untreated age-matched male chicks from the same batch served as controls. After decapitation of the chicks, the eyes were enucleated and the retina was prepared. The retinae from both eyes of each chick were pooled for RNA isolation. Experiment Overall Design: Four samples each: Retina of both eyes of +6.9 diopter lens-treated chicks (plus_lens), Retina of both eyes of untreated control chicks (control)
