Project description:Little is known about cell responses and survival in the presence if reducing agents that could change the redox state of living organisms potentially conferring a negative effect on major metabolic pathways and respiration. As many reducing reagents, in particular sulfur-containing compounds, are widely used as food preservatives, we decided to study effects of sublethal concentrations of reducing reagents on bacterial adaptation. The aim of this study was to analyse and compare the transcriptome profils of untreated and sulfite-treated Escherichia coli cells obtained by employing microarray analysis.

Project description:Little is known about cell survival in the presence of reducing reagents, likewise changing the redox state of living organisms and, therefore potentially conferring a negative effect on respiration and major metabolic pathways. We employed Escherichia coli as a model organism to analyze its responses to sublethal concentrations of sodium sulfite, one of the common food preservatives used for inhibition of microbial growth. We found that partial inhibition of E. coli growth by sodium sulfite was accompanied by profound changes in gene expression. Besides affecting major metabolic pathways, these changes apparently led to reduced production of flagellar proteins, thus potentially impacting cell motility. Concomitantly, E. coli upregulated a number of genes involved in repair of macromolecules and antisense control.

Project description:Purpose: In order to clarify the molecular response mechanism of Tetranychus truncatus to sublethal concentrations of abamectin. Methods: We sequenced the transcriptome of Tetranychus truncatus treated with sublethal concentration of abamectin and control. Results: we provide a comprehensive transcriptome resource for Tetranychus truncatus derived from treating with sublethal concentration abamectin and control. Conclusions: The new genomic resources and data that we present in this study for Tetranychus truncatus will substantially facilitate molecular studies of underlying mechanisms involved in abamectin resistance.

With a disruption of the epidermal barrier, water is lost resulting in an increase in extracellular sodium concentration. We demonstrate that the sodium channel Nax functions as the sodium sensor. With increased extracellular sodium, Nax up-regulates prostasin which results in activation of the sodium channel ENaC, resulting in increased sodium flux and increased downstream mRNA synthesis of inflammatory mediators. The same pathways are present in lung epithelial cells. A signal transduction pathway mediated directly through Nax and secondarily through ENaC results in production of secretory inflammatory mediators. These mediators result in epithelial proliferation and restoration of epidermal homeostasis, but can also have negative effects including excess inflammation and ultimately leads to activation of fibroblasts.

Project description:Despite the widespread use of antiseptics such as chlorhexidine digluconate (CHX) in dental practice and oral care, the risks of potential resistance toward antiseptics in oral bacteria have only been highlighted very recently. Since the molecular mechanisms behind antiseptic resistance or adaptation are not entirely clear yet and the bacterial stress response has not been investigated systematically so far, the aim of the present study was to investigate the transcriptomic stress response in Streptococcus mutans after treatment with CHX using RNA sequencing. Planktonic cultures of stationary phase S. mutans were treated with a sublethal dose of CHX (125 µg/mL) for 5 min. After treatment, RNA was extracted, and RNA sequencing was performed on the Illumina NextSeq 500. Differential expressed genes were analyzed and validated by qRT-PCR. The analysis of the differential gene expression following pathway analysis revealed a considerable number of genes and pathways significantly regulated in S. mutans after sublethal treatment with CHX. In summary, expression of 423 genes was up-regulated and 295 genes down-regulated after CHX treatment. Analysis of differentially expressed genes and significantly regulated pathways showed regulation of genes involved in purine nucleotide synthesis, biofilm formation, transport systems and stress responses. In conclusion, these results show an overview of the transcriptomic stress response in S. mutans upon exposure to CHX and give an insight in potential mechanisms that may result in development of resistances.

Both wild type and Nax knockdown HaCaT cells were seeded in 12-well plates. 70-80% confluent cells were starved with fetal bovine serum (FBS) free Dulbecco's modiM-oM-,M-^Aed Eagle's medium (DMEM) overnight followed by a treatment with control medium (final150 mM sodium) or medium plus 10% more sodium (final 165 mM sodium). Cells were harvested at time point 0, 4, and 16 hours post treatment.

Project description:Due to the recurring loss of antimalarial drugs to resistance, there is a need for novel targets, drugs, and combination therapies to ensure the availability of current and future countermeasures. Pyrazoleamides belong to a novel class of antimalarial drugs that disrupt sodium ion homeostasis, although the exact consequences of this disruption in Plasmodium falciparum remain under investigation. In vitro experiments demonstrated that parasites carrying mutations in the metabolic enzyme PfATP4 develop resistance to pyrazoleamide compounds. However, the underlying mechanisms that allow mutant parasites to evade pyrazoleamide treatment are unclear. Here, we first performed experiments to identify the sublethal dose of a pyrazoleamide compound (PA21A092) that caused an approximately 50% reduction in growth over one intraerythrocytic developmental cycle (IDC). At this drug concentration, we collected transcriptomic and metabolomic data at multiple time points during the IDC to quantify gene- and metabolite-level alterations in the treated parasites. To probe the effects of pyrazoleamide treatment on parasite metabolism, we coupled the time-resolved omics data with a metabolic network model of P. falciparum. We found that the drug-treated parasites adjusted carbohydrate metabolism to enhance synthesis of myoinositol—a precursor for phosphatidylinositol biosynthesis. This metabolic adaptation caused a decrease in metabolite flux through the pentose phosphate pathway, causing a decreased rate of RNA synthesis and an increase in oxidative stress. Our model analyses suggest that downstream consequences of enhanced myoinositol synthesis may underlie adjustments that could lead to resistance emergence in P. falciparum exposed to a sublethal dose of a pyrazoleamide drug.
