Project description:Microglia cells are the innate immune cells of the brain. They are increasingly studied for their role in virtually all pathological events in the central nervous system. However, no study has thus far looked at whether microglia cells undergo wake and sleep state-dependent changes in transcriptional and translational profile. Rodents are nocturnal animals, and most studies are investigating microglia cells during the rodent's sleep phase, hence during the day, for translational purposes. The aim of this study was to compare the microglial transcriptional, translational and functional profile between the wake and sleep phase. Our results provide evidence that the innate immune cells of the brain do undergo a transcriptional and translational adaptation to the wake or sleep state.

Profiling molecular heterogeneity in human primary microglia

Project description:In the brain the cells that control inflammation are called a type of white blood cell called microglia. Microglia are located throughout the brain and spinal cord and account for 10Â–15% of all cells found within the brain. As the resident white blood cells, they are the main active immune defence in the central nervous system (CNS). Microglia are part of an important class of cells known as macrophages that have two main states: M1 and M2. M1 cells are pro- inflammatory, leading to more inflammation, while M2 are anti-inflammatory, and drive wound healing. In this study, we will collect primary microglia from surgical biospies of 100 individuals. This data is part of a pre-publication release. Proteome profiling of LMNA-deficiency microglia activation

Project description:To study the role of LMNA gene in the central nervous system, LMNA gene knock-out mice model and BV2 cell line were obtained. Microglia activation was detected in mice and BV2 cell lines. Phagocytic activity and cytokine production were determined with immunofluorescent and qPCR respectively. Protein profiling of primary microglia and BV2 cells was identified with LC-MS/MS. Differentially expressed proteins were validated by western blotting.
