Project description:Spermatogenesis is a highly regulated process that produces sperm which faithfully transmits genetic information to the next generation. Although extensively studied in mice, our current understanding of human and macaque spermatogenes is limited to known populations defined by state-specific markers developed from rodent data. As between-species differences have been reported in the process duration and cellular differentiation hierarchy, it remains unclear how molecular markersand cell states are conserved or have diverged from mice toman. To address this challenge, we employ single-cell RNA sequencing to identify transcriptional signatures of major germ and somatic cell-types of the testes in both human and macaques, and compared these data with mouse to characterize conserved and divergent features. This approach reveals differences in gene regulation throughout spermatogenesis, including differences in the stem/progenitor pool of spermatogonia, the expression timing of classical markers of differentiation, potential regulators of meiosis, the kinetics of RNA turnover during sperm maturation, and germ cell-soma communication. These datasets provide a rich foundation for future targeted mechanistic studies of germ cell development and in vitro gametogenesis.

Project description:Small RNA including microRNA (miRNA) and Piwi-interacting RNA (piRNA) play important roles in germline maintenance and maturation in wide variety of species. Relatively little however is known about role played by miRNA in male germline maturation in humans and closely related primate species. Here we focused on rhesus macaques as a model species closely related to humans to study small RNA expression in testis samples throughout postnatal development and maturation. We observe clear transition in miRNA and piRNA expression resulting in the overall increase of piRNA expression levels and corresponding decrease in miRNA expression. Unexpectedly, this transition takes place at approximately one year of age – far earlier then rhesus macaque sexual maturation occurring at 4-5 years of age. Notably, contradictory to the overall trend of expression decline, a group of 29 miRNA showed marked expression increase in macaque testis at approximately five years of age – the time interval associated with sexual maturation. Project description:Aging of population is a great challenge of healthcare. In china, the number of the elderly is rapidly growing, and it was estimated that there will be approximately 400 million citizens above 65 years old in 2050.Study on the changes of brain during aging may help elucidate the mechanism of the pathological process, and hence prevent or treat these neurological diseases.Rhesus macaque (Macaca mulatta) and human have a genetic homology of 95%, and their anatomy structures or physiological process are highly similar, which make rhesus macaque one of the most important nonhuman primate models.Thus, the comparison between the change of protein profile during aging in human and rhesus macaque is still necessary, and the characteristics of proteins that are conservative or divergent are of interest.The aim of the(our) study is to identify the conservative changes of pathways during aging, and to reveal the potential difference between human and rhesus macaque so that relevant studies based on primate models can be interpreted more accurately.

Project description:Aging of the human testis and associated cellular changes are difficult to assess, thus we used a translational non-human primate model to get insights into underlying biochemical processes. Using proteomics we analyzed testicular tissue of six young (age 2 to 3) and four old (age 10 to 12) C. jacchus individuals. Mass spectrometry identified 63124 peptides, which could be assigned to 5924 proteins. Among them we found proteins, which are specific for germ cells, Leydig cells, and somatic cells, indicating the relevance of the dataset. The quantitative analysis showed 31 differentially abundant proteins, of which 29 proteins were more abundant in old animals. An increased abundance of several anti-proliferative proteins, among them CDKN2A, indicate slowed cell proliferation in older testes. Immunohistochemistry localized CDKN2A in spermatogonia and spermatocytes, suggesting altered spermatogenesis. Additionally, an increased abundance of several extracellular proteins and small leucine rich repeat proteoglycans was observed, which may be related to impaired cell migration and fibrotic events. An increased abundance of proteins with inhibitory roles in smooth muscle cell contraction like CNN1, indicate functional alterations, specifically in peritubular cells and may mirror a reduced capacity of these cells, to contract in aged testes. In summary, these results document several age-associated changes in the testicular proteome of a non-human primate model and provides new insight into testicular aging.

Project description:Primordial germ cells (PGCs) are the earliest embryonic progenitors in the germline. Correct formation of PGCs is critical to reproductive health as an adult. Recent work has shown that primate PGCs can be differentiated from pluripotent stem cells; however, a bioassay that supports their identity as transplantable germ cells has not been reported. Here, we adopted a xenotransplantation assay by transplanting single-cell suspensions of human and nonhuman primate embryonic Macaca mulatta (rhesus macaque) testes containing PGCs into the seminiferous tubules of adult busulfan-treated nude mice. We discovered that both human and nonhuman primate embryonic testis are xenotransplantable, generating colonies while not generating tumors. Taken together, this work provides two critical references (molecular and functional) for defining transplantable primate PGCs. These results provide a blueprint for differentiating pluripotent stem cells to transplantable PGC-like cells in a species that is amenable to transplantation and fertility studies.

To determine the dynamics of active enhancers during spermatogenesis, we performed H3K27ac ChIP-seq and detected reagions of active enhancers during spermatogenesis. We analyzed four representative stages of spermatogenesis: Thy1+ undifferentiated spermatogonia, which contains spermatogonial stem cells and progenitor cells; c-Kit+ differentiating spermatogonia from P7 testes; purified pachytene spermatocytes (PS) undergoing meiosis; and postmeiotic round spermatids (RS) from adult testes.
