Project description:Chronic hypertension during pregnancy affects both mother and infant but its pathological mechanisms are unclear. Due to ethical limitations human data is available in term tissue only- so deeper understanding of the vascular changes during early pregnancy are unknown. This study utilises a known rat model of chronic hypertension to study changes in gene expression at an early gestational time point. After 6 days of pregnancy animals were sacrificed and main uterine arteries harvested for RNA-Seq (or age-matched virgin rats as controls).

Transcriptome profile form endometrial biopsy samples of pregnant and non-pregnant cows

Project description:Purpose: characterize the uterine transcriptome profiles of pregnant (P) versus non-pregnant (NP) cows during early pregnancy and attempted to define a potential set of marker genes that can be valuable for predicting pregnancy outcome. Methods: beef cows were synchronized and artificially inseminated at detected estrus. Six days after AI, jugular blood samples and a biopsy from the uterine horn contralateral to the ovary containing the corpus luteum were collected. Based on pregnancy outcome on day 30, samples were retrospectively allocated to the following groups: Pregnant and Non-Pregnant. Both groups had similar plasma progesterone concentrations on D6. Uterine biopsies were submitted to RNA-Seq analysis in a Illumina HiScanSQ platform. Results: The 272,685,768 million filtered reads were mapped to the Bos taurus reference genome and 14,654 genes were analyzed for differential expression between groups. Transcriptome data showed that 216 genes are differently expressed when comparing NP versus P uterine tissue (Padj≤0.1). More specifically, 36 genes were up-regulated in P cows and 180 are up-regulated in NP cows. Conclusions: this study characterized a unique set of genes, expressed in the uterus on 6 days after insemination, that indicate a receptive state leading to pregnancy success. Whole-genome transcriptome profiling in PFOS-treated uterine artery endothelial cells isolated from pregnant women

Project description:During pregnancy, perfluorooctane sulfonate (PFOS) exposure is linked to increased risks of preeclampsia and fetal developmental complications. Although experimental and circumstantial data suggest that PFOS induces endothelial dysfunction, leading to decreased uterine arterial blood flow and gestational hypertension, the precise regulatory mechanisms responsible for this effect remain unknown. To address this issue, we treated human uterine artery endothelial cells (hUAECs) isolated from pregnant women with PFOS and conducted a comparative transcriptomic analyses to understand the underlying mechanism of PFOS-induced endothelial dysfunction.

Whole-Genome Uterine Artery Transcriptome Profiling and Alternative Splicing Analysis in Rat Pregnancy.

Project description:During pregnancy, the uterine artery (UA) undergoes extensive remodeling to permit a 20-40 fold increase in blood flow with associated changes in the expression of a multitude of genes. This study used next-gen RNA sequencing technology to identify pathways and genes potentially involved in arterial adaptations in pregnant rat UA (gestation day 20) compared with non-pregnant rat UA (diestrus). A total of 2245 genes were differentially expressed, with 1257 up-regulated and 970 down-regulated in pregnant UA. Gene clustering analysis revealed a unique cluster of suppressed genes implicated in calcium signaling pathway and vascular smooth muscle contraction in pregnant UA. Transcription factor binding site motif scanning identified C2H2 ZF, AP-2 and CxxC as likely factors functional on the promoters of down-regulated genes involved in calcium signaling and vascular smooth muscle contraction. In addition, 1686 genes exhibited alternative splicing that were mainly implicated in microtubule organization and smooth muscle contraction. Cross-comparison analysis identified novel genes that were both differentially expressed and alternatively spliced; these were involved in leukocyte and B cell biology and lipid metabolism. In conclusion, this first comprehensive study provides a valuable resource for understanding the molecular mechanism underlying gestational uterine arterial adaptations during pregnancy.
