Transcriptional signature of murine tracheal brush cells identified by RNA-Seq

Project description:Purpose: Tracheal epithelial brush cells are rare chemosensory cells defined by their expression of elements of the bitter taste transduction system, and known to activate the cholinergic nervous system in the murine lung. Similar chemosensory cells in the intestine can generate lipid mediators and pro-inflammatory cytokines but whether brush cell can contribute to airway inflammation is unknown. Furthermore, despite the advances in understanding chemosensory cell effector functions, the receptors that mediate chemosensory cell activation and expansion beyond taste receptors in any compartment remain largely unknown. Methods: In this study, we isolated tracheal brush cells by FACS from naïve ChATBAC-eGFP mice with knockin of eGFP within a BAC spanning the acetylcholine transferase locus, marking brush cells in the epithelium and performed transcriptome profiling using low input RNA sequencing. We compared tracheal brush cells to EpCAM+ epithelial cells and CD45+ hematopoetic cells in naive mice. Results: When compared to EpCAM+ EpCs and to CD45+ cells in the airway, principal component analysis demonstrated that brush cells grouped quite distinctly. This brush cell distinction relative to EpCAM+ cells, was further reflected in the striking number of highly differentially expressed genes. This included 1305 genes expressed at 4-fold or higher levels in EpCAM+eGFP+ cells (brush cells), of which 418 genes were expressed at 32-fold or higher levels in brush cells. Conclusions: Our study represents the first detailed analysis of the transcriptome of tracheal brush cells and identifies a unique set of genes that are primarily expressed in brush cells including the bitter taste transduction system, synthenic machinery for several pro-inflammatory lipid mediators and HoxA2 transciptional factors.

Gene expression analysis of human fallopian tube stem cells and air-liquid interface differentiation

Project description:If the fallopian tube is the origin of serous cancer, one possible mechanism for the evolution of cancer is a dysregulation of indigenous stem cells. We therefore set out to clone the stem cells of the human fallopian tube using methods to clone columnar epithelial stem cells such as human intestinal stem cells. Using this method, we were able to generate clones of fallopian tube stem cells that contain many small, undifferentiated cells. These stem cell clones show strong and consistent staining with markers of fallopian tube epithelial cells (PAX8). We also established an air-liquid interface culture system to differentiate fallopian tube stem cell to both ciliated cells and non-ciliated cells.

SARS-CoV-2-infected human nasal epithelial air-liquid interface cultures

Project description:Continuous assessment of the impact of SARS-CoV-2 on the host at the cell-type level is crucial for understanding key mechanisms involved in host defense responses to viral infection. We investigated host response to ancestral-strain and Alpha-variant SARS-CoV-2 infections within air-liquid-interface human nasal epithelial cells from younger adults (26-32 Y) and older children (12-14 Y) using single-cell RNA-sequencing. Ciliated and secretory-ciliated cells formed the majority of highly infected cell-types, where the latter derived from ciliated lineages. Strong innate immune responses were observed across lowly-infected and un-infected bystander cells and heightened in Alpha-infection. Alpha highly-infected cells showed increased expression of protein-refolding genes compared with ancestral-strain-infected cells in children. Furthermore, oxidative phosphorylation-related genes were down-regulated in bystander cells versus infected and mock-control, underscoring the importance of these biological functions for viral replication. Overall, this study highlights the complexity of cell-type-, age- and viral strain-dependent host epithelial responses to SARS-CoV-2.

Transcriptional profiling by array to investigate the effect of interfacial stress on rat alveolar type II cell signalling and gene expression (conventional air-liquid interface)

Project description:Many epithelia have contact with air-liquid interfaces. This applies particularly to the lung, where one of the epithelial cell types (the surfactant secreting alveolar type II [AT II] cells) even project into the air. This specific environment may be of considerable physiological relevance; however, only few data exist to provide a satisfying description. This is mainly due to the difficulty to manipulate cell-air contacts in a specific way. In previous investigations, using new microscopic approaches, we found that the presence of an air-liquid interface leads to a paradoxical situation: it is a potential threat that may cause cell injury, but also a potent stimulus for the cells: AT II cells respond promptly, and show sustained Ca2+-signals that activate exocytosis. In this report, gene chip analysis was performed to test the concept of an Air-liquid interface IAL as a mechanical stimulus, stress factor and/or regulator of the AT II cells. For this purpose, cultured AT II cells were exposed to two different interfacial conditions (inverted and conventional interface). The results support the concept that the IAL is a strong stimulus for AT II cells inducing cellular responses that seem to be closely associated with mechanotransduction, stress response, and alveolar repair.
