Project description:3D cell culture models are recognized for representing the physiological microenvironment and exhibiting higher concordance with in vivo conditions, when compared to a conventional 2D cell culture model. However, cells grown in 3D cultures are likely to exhibit slower growth than those in 2D cultures. We found that addition of a novel small molecule named GA-017 to culture media promotes the cell proliferation particularly under 3D conditions. Gene microarrays were used to observe the global gene expression in Skov3 cells cultured under 3D condition with DMSO or GA-017 and identified distinct classes of up or down-regulated genes.

We treated SKOV3 cells with 5% trehalose (control group) and rhCCL20 protein dissolved in 5% trehalose (experimental group) in vitro. The cells were collected 24 hours after treatment and used for RNA-sequencing.

RNA Sequencing facilitates quantitative analysis of differently expressed genes with GA or SKN treatment in rheumatoid fibroblast-like synoviocytes

Project description:To explore downstream gene expression profile of PKM2,we evaluated the transcriptome of GA- or SKN-treated RA FLSs compared to untreated control using RNA sequencing analysis. The data were analyzed for differential expression with a nominal P<0.05. GA Treatment induced downregulation of 46 genes and upregulation of 58 genes compared to DMSO treatment. SKN treatment resulted in downregulation of 14 genes and upregulation of 27 genes compared to DMSO treatment.A Venn diagram showed the overlap of the significantly expressed genes between GA and SKN treatment. We found that in the overlap of both GA and SKN treatment, STAT5A and ARHGAP11A were significantly downregulated, and THAP6 and AHRR were significantly upregulated.
