Project description:RNA extracted from representative isolates of each serovar grown to late exponential phase in Luria-Bertani broth showed that transcript abundances of core genes were significantly higher (p < 0.05) than those of accessory genes for all three serovars. Inter-serovar comparisons demonstrated higher transcript abundances for genes related to vitamin B12 biosynthesis, and ethanolamine and 1,2-propanediol utilization in S. Cerro compared to Javiana and Typhimurium, while genes in Salmonella Pathogenicity Island (SPI) 1 had significantly higher transcript abundances in S. Javiana and S. Typhimurium.

Proteomic comparison between two Salmonella serovars with differential pathogenicity in humans.

Project description:Salmonella infections are among the most common foodborne diseases worldwide. The Enteritidis and Dublin serovars of Salmonella enterica are closely related yet they differ significantly in pathogenicity and epidemiology. Enteritidis is a broad-host-range serovar that commonly causes gastroenteritis and infrequently causes invasive disease in humans. Dublin mainly colonizes cattle but upon infecting humans often results in invasive disease. The aim of this work was to elucidate the molecular factors responsible for the differential pathogenic behavior between both serovars. We performed a quantitative proteomic comparative analysis between one clinical isolate of each serovar grown in vitro under gut mimicking conditions (GMC). Compared to S. Enteritidis, the S. Dublin proteome was enriched in proteins linked to response to several stress conditions, such as those encountered during host infection, as well as to virulence. The S. Enteritidis proteome contained proteins related to central anaerobic metabolism pathways that were undetected in S. Dublin. Similar differences were also found at the transcriptional level, as mRNA levels correlated with proteomic results for 17 of the 20 genes tested in 4 natural isolates of each serovar grown in GMC. This work reveals proteomic differences between two Salmonella serovars with markedly different invasive and host-range characteristics, grown in an infection relevant condition, which were not evident in previous comparative genomic analyses.

Project description:The objective of the present investigation was to consider the level of variation in the protein expression patterns of closely related Salmonella serovars, in order to search for protein factors with levels of expression or posttranslational modifications characteristic for each serovar. For the comparative expression analysis we have utilised classic 2D GE approach which revealed several proteins with serovar specific expression as well as proteins which do not alter their expression levels between serovars and strains. Genomic comparison of Salmonella enterica serovars and Salmonella bongori

Project description:This experiment set includes 64 arrays representing 26 serovars and strains of Salmonella spp. including many representatives of subspecies I, Arizona from subsp. IIIa, and S. bongori from subsp. V. The genomic DNA from all strains were labeled with Cy5 and hybridized against an equal amount (1.5 ug) of S. typhimurium SL1344 reference genomic DNA that was labeled with Cy3, all on an S. typhimurium SL1344 spotted DNA microarray. Most of the arrays are present in triplicate to account for variability in probe generation, hybridization, and slide quality. Several are represented in duplicate, and a few without any replicates. Single-cell RNA sequencing analysis reveals Greater Epithelial Ridge cells degeneration during postnatal development of cochlea in Rats

Project description:Greater epithelial ridge cells, a transient neonatal cell group in the cochlear duct, which plays a crucial role in the functional maturation of hair cell, structural development of tectorial membrane, and refinement of audio localization before hearing. Greater epithelial ridge cells are methodologically homogeneous, while whether different cell subtypes are existence in this intriguing region and the degeneration mechanism during postnatal cochlear development are poorly understood. In the present study, single-cell RNA sequencing was performed on the cochlear duct of postnatal rats at day 1 (P1) and day 7 (P7) to identify subsets of greater epithelial ridge cell and progression. Gene ontology and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis were used to examine genes enriched biological processes in these clusters. We identified a total of 26 clusters at P1 and P7 rats and found that the cell number of five cell clusters decreased significantly, while four clusters had similar gene expression patterns and biological properties. The genes of these four cell populations were mainly enriched in Ribosome and P13K-Akt signal pathway. Among them, Rps16, Rpsa, Col4a2, Col6a2, Ctsk, and Jun are particularly interesting as their expression might contribute to the greater epithelial ridge cells degeneration. In conclusion, our study provides an important reference resource of greater epithelial ridge cells landscape and mechanism insights for further understanding greater epithelial ridge cells degeneration during postnatal rat cochlear development
