Project description:We investigated antibacterial activities of benzyl isothiocyanate (BITC) against V. parahaemolyticus by transcriptomic analysis and morphological verification. Treatment with 1/8MIC BITC resulted in 234 up-regulated genes and 273 down-regulated genes. In V. parahaemolyticus, we selected six genes that were significantly down-regulated from the transcriptome results and verified them using quantitative Real-time Polymerase Chain Reaction (qRT-PCR). The verification experiments indicated that the relative expressions of the six virulence genes VP0820, VP0548, VP2233, VPA2362, fliA and fliG were only 31.0%, 31.1%, 55.8%, 57.0%, 75.3%, and 79.9% of the control group, respectively. Among them, VP2233, fliA and fliG are related to flagella, and VP2362 can regulate a protein related to biofilm formation. From the morphological aspect, we verified that under the action of 1/8 MIC BITC, the swimming diffusion diameter of V. parahaemolyticus was significantly reduced by 14.9%, and biofilm formation was significantly inhibited. These results indicate that 1/8 MIC BITC can reduce the pathogenicity of V. parahaemolyticus by inhibiting virulence gene expression related to flagella and biofilm, which is helpful for further understanding of bacteriostatic mechanism of BITC on V. parahaemolyticus and other food-borne pathogens.

Project description:BackgroundFoodborne illness caused by Vibrio parahaemolyticus (V. parahaemolyticus) is generally associated with the consumption of seafood. Fish and other seafood can be contaminated with V. parahaemolyticus, natural inhabitants of the marine, estuarine, and freshwater environment. In this study, the antibacterial activities of benzyl isothiocyanate (BITC) against V. parahaemolyticus were investigated by both transcriptomic analysis and morphological verification.ResultsTreatment with 1/8 minimum inhibitory concentration (1/8 MIC) BITC resulted in 234 upregulated genes and 273 downregulated genes. The results validated by quantitative real-time polymerase chain reaction (qRT-PCR) revealed that the relative expression levels of the six genes VP0820, VP0548, VP2233, VPA2362, fliA and fliG were only 31.0%, 31.1%, 55.8%, 57.0%, 75.3%, and 79.9% of the control group, respectively. Among them, genes VP2233, fliA and fliG are related to flagella and VP2362 can regulate a protein relevant to biofilm formation. Morphologically, we verified that the swimming diffusion diameter of V. parahaemolyticus was significantly reduced by 14.9% by bacterial swimming ability, and biofilm formation was significantly inhibited by treatment with 1/8 MIC BITC by crystal violet quantification assay.ConclusionsThese results indicated that 1/8 MIC BITC had antibacterial effect on V. parahaemolyticus by inhibiting virulence gene expression related to flagella and biofilm.

Transcriptome analysis of different concentrations of benzyl isothiocyanate against Staphylococcus aureus

Project description:In this study, we monitored the transcriptional changes of S. aureus by RNA-seq analysis to better understand the effect of benzyl isothiocyanate (BITC) on the virulence inhibition of S. aureus and determined the bacteriostatic effect of BITC at subinhibitory concentrations. Our results revealed that compared with the control group (SAC), the BITC-treated experimental group (SAQ_BITC) had 708 differentially expressed genes (DEGs), of which 333 genes were downregulated and the capsular polysaccharide (cp) was significantly downregulated. Furthermore, we screened five of the most virulent factors of S. aureus, including the capsular polysaccharide biosynthesis protein (cp5D), capsular polysaccharide synthesis enzyme (cp8F), thermonuclease (nuc), clumping factor (clf) and protein A (spa).

Project description:Allyl isothiocyanate (AITC) is a natural product used as a food additive. Due to its strong volatility and broad biological activity, AITC is being considered as a bio-fumigant to control soil-borne fungal diseases in agriculture, creating an urgent need for evaluation of the antifungal activity of AITC. Here, AITC-trigged growth inhibition, sensing, and related molecular mechanisms were analyzed in Fusarium solani by employing morphological, genetic, and transcriptional profiling analysis. The results indicated that AITC causes rapid inhibition of F. solani after 5 min, hyphal deformity, and electrolyte leakage. A yeast-like vacuolar transient receptor potential channel regulator (FsYvc1, a STRPC family member) was identified in F. solani that is play a switch role for sensing AITC. Genetic evidence suggests the gene FsYvc1 involved in growth, development, and pathogenicity. Loss of FsYvc1 resulted in hypersensitivity of F. solani to AITC and high levels of reactive oxygen species with weak responses to CaCl2, NaCl, KCl, SDS, and Congo red as well as triadimefon, triadimenol, chlorothalonil, and azoxystrobin. Transcription profiling and qRT-PCR showed that three transcripts were up-regulated in the wild type (WT) but down-regulated in the FsYvc1 mutants (∆FsYvc1), which were involved in TRP ion channel expression, biosynthetic process, and oxidoreductase activity. Three transcripts down-regulated in WT but up-regulated in the ∆FsYvc1 were involved in metabolic process, proper enzyme folding (GrpB protein), and the response to abiotic stress (WD40 repeat). This study provides novel insights into the toxicity mechanisms of AITC. Considering the key role of FsYvc1, it could act as a new molecular target for future fungicide development.

Project description:Vibrio parahaemolyticus is a Gram-negative marine bacterium. Strain RIMD 2210633, the wild type strain of the organism, causes acute gastroenteritis in humans. Human intestinal factor bile often affects the global gene regulation in some species of enteropathogenic bacteria. To determine the genes in V. parahaemolyticus that respond to bile, we investigated the differences in the transcriptomes of the wild type strain and the vtrA-null strain grown in Luria-Bertani medium cultivated with or without 0.04% crude bile. The vtrA gene encodes the previously identified T3SS2 regulator. Our goal is to demonstrate bile regulon controlled by VtrA in V. parahaemolyticus.
