Biotin-Labelled Clavulanic Acid to Identify Proteins Target for Haptenation in Serum

Project description:Clavulanic acid (CLV) is a betalactam (BL) which inhibits betalactamases activity and is frequently administered combined with amoxicillin (AX). Both BLs can be independently involved in allergic reactions. Indeed, selective immediate allergic reactions to CLV have recently been reported in 30% of patients allergic to the AX-CLV combination. Although protein haptenation with β-lactams is considered necessary to activate the immune system, currently there are no straight-forward detection tools for the study of protein haptenation by CLV. The aim of this study was to assess the suitability of two biotinylated analogues of CLV, CLV-B or CLV-TEG-B (containing the later a hydrophilic tetraethylenglycol linker), as probes to study protein haptenation by this β-lactam. Our results show that tagged CLV keeps some of the features of CLV, as the amide binding by which they haptenate proteins and it could constitute a valuable tool to identify protein targets for haptenation by CLV with high sensitivity to get insights into the activation of the immune system by CLV as well as mechanisms involved in allergy to β-lactams.

Project description:Comparison of the whole genome gene expression level of an amoxicillin resistant E. coli strain with the wildtype it was derived from. The process of amoxicillin adaptation of E. coli MG1655 wildtype cells is further descibed in van der Horst, M, J.M. Schuurmans, M. C. Smid, B. B. Koenders, and B. H. ter Kuile (2011) in Microb. Drug Resist. 17:141-147. Resistance to amoxicillin was induced in E. coli by growth in the presence of stepwise increasing antibiotic concentrations. To investigate consequences of the aquisition of amoxicillin resistance the transcriptomic profile of sensitive and resistant cells was compared in the absence and presence of sub-inhibitory (0.25xMIC) amoxicillin concentrations was compared. Total RNA of 3 biological replicates of E. coli MG1655 wildtype cells and amoxicillin resistant cells cultured with (0.25xMIC) or without amoxicillin was hybridized on a 12x135k custom designed microarraychip against one common reference.

RS07910/07915 TCS multipath regulates clavulanic acid metabolic pathways in Streptomyces clavuligerus F613-1

Project description:Streptomyces clavuligerus F613-1 produces a clinically important β-lactamase inhibitor, clavulanic acid (CA). The biosynthesis pathway of CA has been basically elucidated, however, the global regulation of CA biosynthesis remains unclear. In order to further elucidate the regulatory mechanism of clavulanic acid synthesis, TCS RS07910/07915 which is next to the CA biosynthetic gene cluster was deleted in Streptomyces clavuligerus. F613-1. Deletion of RS07910/07915 results in decreased the production of CA, but the phenotype was not affected. Both the transcriptome and ChIP-seq data revealed that the TCS RS07910/07915 mainly regulate genes involved in primary metabolism (such as fatty acid degradation, glyceraldehyde 3-phosphate (G3P) metabolism, arginine biosynthesis) and CA biosynthesis. EMSA assays revealed that RS07915 could bind to the promoter fragments of argG, argC, oat1, oat2, ceaS1 and claR in vitro, indicating that RS07915 could direct regulate the biosynthesis genes of arginine and CA. This study indicated that RS07910/07915 is a pleiotropy regulatory TCS, RS07910/07915 could directly affect the biosynthesis of CA, and indirectly affect CA production through affecting primary metabolism of arginine and G3P (precursors of CA).
