Alternative culture medium design for biomass production of autochthonous meat starter Latilactobacillus sakei sp. sakei ACU-2.

Project description:The autochthonous strain Latilactobacillus sakei sp. sakei ACU-2 was selected as a meat starter culture for dry sausage production. Transferring this strain from laboratory scale to industry requires an increase in biomass production, while lowering process costs. In this study, a combination of techniques was applied in order to optimize the culture medium composition to enhance biomass production of L. sakei ACU-2. One variable at a time experiments, Plackett-Burman design, and mixture design were performed to fulfill the strain nutritional requirements. Eventually, the optimized formulation contained 19.46 g/L yeast extract; 8.28 g/L whey protein concentrate; 2.26 g/L soy peptone; 30 g/L cerelose; 1 g/L Tween 80; 5 g/L sodium acetate; 0.2 g/L magnesium sulfate and 0.05 g/L manganese sulfate. When L. sakei ACU-2 was cultivated in a bioreactor using the alternative medium, an enhancement of 75.5% of biomass production was achieved, in comparison to its growth in the commercial de Man, Rogosa, and Sharpe medium. Furthermore, a reduction of 62-86% of the cost was also attained. These results support a promising large-scale application of the designed medium for high biomass yields of the starter culture at minor costs.

Project description:Nowadays, given the globalization as well as the numerous technological developments and innovations that govern the food market, consumer’s expectations, regarding the food they eat, have increased and the information they receive should be at least reliable. This proof-of- concept study is the first analysis of potato tuber tissue using integrated multi-omics approaches across genome-wide DNA methylation, RNA sequencing and quantitative proteomics to obtain the molecular portrait of famous PGI potatoes of Naxos island. Furthermore, we used metagenomics analysis in order to discriminate potato tubers produced in diverse PGI regions based on the distinct microbiological patterns. Hence, we reveal key molecular factors related to harvest and post-harvest through the dynamics of causal models.

The effect of the production system on the proteomics profiles of the Longissimus thoracis muscle in Arouquesa cattle

Project description:Arouquesa breed is an autochthonous Portuguese breed produced under a traditional mountain system. It is known for its Arouquesa PDO beef, a high-quality product with valued organoleptic characteristics. There is a need to improve production systems without affecting beef quality. The aim of this work is to compare the proteomics profile from the Longissimus thoracis muscle from Arouquesa animals produced under different systems. Sixty weaners were produced under the following systems: traditional (TF) and traditional with starter feed supplementation (TF+S1) with weaning and slaughtering at 8 months, the S1+S2 (weaning at 5 months and grower supplement until slaughter) and two rearing periods with finishing supplementation (TF+S3 and S3). Upon slaughter, samples of L. thoracis were taken and analyzed using a shotgun proteomics workflow. Several putative biomarkers of beef quality for the Arouquesa breed were identified: VIM, FSCN1, SERPINH1, ALDH1A1, NDUFB5, ANXA1, PDK4, CEMIP2, NDUFB9, PDLIM1, OXCT1, MYH4. These proteins were involved in actin binding, skeletal muscle development and in the mitochondrial respiratory chain and they can influence mostly meat tenderness and color. Our results demonstrate the link between production practices and putative meat quality signatures, that have potential to improve traceability of certified products.
